Terbinafine hydrochloride, although one of the prominent antifungal agents, suffers from low drug permeation owing to its hydrophobic nature. The approach of nanosponge formulation may thus help to resolve this concern. Thus, the present research was envisioned to fabricate the nanosponge hydrogel of terbinafine hydrochloride for topical delivery since nanosponge augments the skin retentivity of the drug. The optimized formulation was obtained using Box Behnken Design. The dependent and independent process parameters were also determined wherein polyvinyl alcohol (%), ethylcellulose (%), and tween 80 (%) were taken as independent process parameters and particle size, polydispersity index (PDI), and entrapment efficiency (EE) were the dependent parameters. The nanosponge was then incorporated into the hydrogel and characterized. In-vitro drug release from the hydrogel was 90.20 ± 0.1% which was higher than the drug suspension and marketed formulation. In vitro permeation potential of the developed formulation through rat skin showed a flux of 0.594 ± 0.22 µg/cm2/h while the permeability coefficient was 0.059 ± 0.022 cm/s. Nanosponge hydrogel was evaluated for non-irritancy and antifungal activity against C. albicans and T. rubrum confirming the substantial outcome. Tape stripping studies exhibited ten times stripping off the skin quantified 85.6 ± 0.21 μg/cm2. The confocal analysis justified the permeation potential of the prepared hydrogel. The mean erythemal score was 0.0, confirming that the prepared hydrogel did not cause erythema or oedema. Therefore, based on results obtained, nanosponge hydrogel formulation is a potential carrier for efficient topical delivery of terbinafine hydrochloride.

BACKGROUND Although the structure and physicochemical properties of large ring cyclodextrins (LR‐CDs) exhibit unique characteristics, and also possess very strong water solubility and high safety, little is known about the embedding performance of macrocyclodextrin. Encapsulation refers to a complex of tea tree oil (TTO) with the wall material, protecting the core material or changing its properties from adverse external factors, controlling its release rate against the evaporation and degradation of essential oils. In the present study, LR‐CDs complexed with TTO were prepared by co‐precipitation methods. RESULTS The mass ratio of LR‐CDs‐TTO was six and the maximum complexation efficiency was 86.23%. Fourier‐transform infrared spectroscopy analysis presented the loss of characteristic peaks related to TTO in the complex and no other additional peaks were observed. X‐ray diffraction examination demonstrated several sharp peaks and intensity peaks at the diffraction angle of the TTO‐LR‐CDs complex. 1H‐NMR indicated a chemical shift as a result of the interaction between the molecules in the inclusion complex. Moreover, the thermal stability and aqueous solubility of TTO were enhanced after synergy with LR‐CDs; particularly, the solubility of the complex was increased by 329‐fold. The volatile characteristics of the encapsulated and original TTO were identical. CONCLUSION The results of the present study show that TTO was efficaciously complexed with LR‐CDs and exhibited enhanced solubility and thermal stability. © 2020 Society of Chemical Industry

This work aims to optimize a new formulation of orange nectar sweetened with Stevia and enriched with Pectin using full factorial design with two factors, two levels, and one central point. Factors were the concentration in stevia and pectin. Physicochemical characterization and stability study were carried‐out at 4 and 25°C where pH, Brix°, total titratable acidity, turbidity, sedimentation, and redispersibility are controlled for 1 month. The evaluation of antioxidant activity degradation during 3 months and organoleptic test were required for the selection of optimal formulation. The hypoglycemic effect of the optimal nectar was evaluated in rats targeting the postprandial blood glucose levels kinetics. After optimization, the formulation F1 (with 0.03% stevia and 0.05% pectin) was found to be the most stable and favorable. Its half‐life times were, respectively, 147.67 days at 4°C (R2 = .996) and 74.66 days at 25°C (R2 =.997). Its organoleptic score was 9.67 (R2 = .994). The in‐vivo hypoglycemic study has been confirmed, the effect of this supplementation has permitted the regulation of post‐prandial glycemic levels. Thus, this formulation deserves to be recommended for diabetic subjects. Practical applications The addition of stevia (0.03%) and pectin (0.05%) improved the physico‐chemical stability of natural orange nectar. Referring to the antioxidant activity degradation; half‐life time was 147.67 days at 4°C and 74.66 days at 25°C. The consumption of this orange nectar by diabetic patients allows the normalization of their postprandial blood glucose levels.
