“…We propose that this procedure is more relevant to screen than more traditional procedures that assess the effects of medications when non-quitting smokers have unrestricted access to smoking Perkins et al, 1992;Dawe et al, 1995;Benowitz et al, 1998;Brauer et al, 1999;Caskey et al, 1999;Jarvik et al, 2000;King and Meyer, 2000;Cousins et al, 2001;Houtsmuller et al, 2002). The main advantage to the present paradigm is that non-quitting smokers are forced to make an ''either/or'' choice between the cigarette and money, a situation that models the choice made by treatment-seekers who are motivated by intrinsic factors to quit smoking.…”

“…Studying effect of cigarettes in treatment seekers is constrained on the ethical grounds; therefore, many prior studies have modeled tobacco addiction by permitting unrestricted access to smoking in nonquitting smokers Perkins et al, 1992;Dawe et al, 1995;Benowitz et al, 1998;Brauer et al, 1999;Caskey et al, 1999Caskey et al, , 2002Jarvik et al, 2000;Cousins et al, 2001;Houtsmuller et al, 2002). However, this design may lack the sensitivity to detect weaker medication effects.…”

“…Chronic nicotine has been shown to increase m-OR in striatum in female rats and in the ventral tegmental area in male and female mice (Walters et al, 2005;Wewers et al, 1999). The opiate antagonist naloxone can block the subjective effects of nicotine in chronic smokers (Brauer et al, 1999;King and Meyer, 2000). Thus, it is not surprising that naltrexone has been proposed for the treatment of nicotine dependence (Schnoll and Lerman, 2006).…”

“…In another recent acute dose study, naltrexone significantly reduced the total number of cigarettes smoked and expired carbon monoxide levels compared to placebo (Epstein and King 2004). Naltrexone administered orally for 1 week increased the aversive effects of the first cigarette smoked each day and attenuated the increased wakefulness after smoking without altering ad lib smoking in the laboratory setting (Brauer et al 1999). Taken together, these studies suggest that naltrexone may influence some aspects of cigarette smoking and not others.…”
