N Protein of Festuca Leaf Streak Virus (Rhabdoviridae) Detected in Cytoplasmic Viroplasms by Immunogold Labelling

Abstract: Ultrathin sections of plant tissue infected with Festuca leaf streak virus (FLSV) were treated with polyclonal antisera against the three major structural proteins, G, N, or M, followed by protein A‐gold labelling. At the periphery of viroplasms, comprising groups of virions, all three major structural proteins were detected. In the granular matrix of the cytoplasmic viroplasms only N protein was detected. “…1979). FLSV N-protein is present in the viroplasms (Lundsgaard, 1992) and Gprotein can be detected in the ER (Lundsgaard, 1995) of infected cells, thus supporting the concept that viroplasms are the site for viral nucleocapsid synthesis with subsequent virion maturation at the surrounding ER membranes.…”

“…Production of the antiserum against the FLSV G and N proteins (ami-FLSV-G and anti-FLSV-N) used in the present study has been described by Lundsgaard (! 992), Anti-FLSV-G did not cross-react with FLSV M or N protein when tested by Western immunoblotting of purified FLSV (Lundsgaard, 1992), but cross-reacted with healthy leaf tissue of F. gigantea in immunogoid labelling control experiments.…”

“…Among the subgroup 1 viruses, barley yellow striate mosaic virus (BYSMV), festuca leaf streak virus (ELSV), and northern cereal mosaic virus (NCMV) induce viroplasms in the cytoplasm (Conti and Appiano, 1973;Toriyama, 1976;Lundsgaard and Albrechtsen, 1979). The viroplasm contains viral N protein and is probably the site for viral transcription and replication (Lundsgaard, 1992).…”
