N-Glycomic Changes in Serum Proteins During Human Aging

Abstract: N-glycan profiling of the human serum glycoproteins including immunoglobulin fraction on different age groups of healthy persons shows substantial changes with increasing age in three major N-glycan structures. In individuals more than 40-50 years of age, there is an increase in under-galactosylated glycans and a decrease in the core alpha-1,6-fucosylated bi-galactosylated biantennary structure. These three glycan structures are also substantially changed in a Werner syndrome patient, to a level comparable or … Show more

“…We previously showed that three major N-glycan structures present in human blood glycoproteins are altered with age (Vanhooren et al, 2007. Here, we investigated the possibility that age-related N-glycosylation changes occur also in mice.…”

“…In our previous studies, we demonstrated that three major N-glycan structures (NGA2F, NGA2FB, and NA2F) present in human blood glycoproteins undergo changes during aging (Vanhooren et al, 2007. These results indicate that the measurement of N-glycan levels could provide a noninvasive surrogate marker for general health, for forecasting disease progression upon aging, and for monitoring the efficacy of antiaging medications.…”

“…Previous studies of suitable size (n Ͼ 200) have performed this to various degrees, finding plasma N-glycans to be highly associated with e.g. age, sex, inflammation, body mass index (BMI), cholesterol and lipid levels (17)(18)(19)(20)(21)(22)(23)(24). However, these studies have either been performed on single proteins, immunoglobulin G (IgG) being a particularly wellstudied example, or predominantly by methods of liquid chromatographic and electrophoretic separation (e.g.…”

“…Moreover, SPRET/Ei mice have attracted interest because they display interesting and robust phenotypes. For instance, they are long-lived (8), and resist the development of multiple forms of cancer or lethal inflammation induced by high doses of LPS or of the cytokine tumor necrosis factor (TNF) (9)(10)(11).…”
