The Methanogens are a diverse group of organisms found in anaerobic environments such as anaerobic sludge digester, wet wood of trees, sewage, rumen, black mud, black sea sediments , etc which utilize carbon dioxide and hydrogen and produce methane. They are nutritionally fastidious anaerobes with the redox potential below -300 mV and usually grow at pH range of 6.0-8.0 [1]. Substrates utilized for growth and methane production include hydrogen, formate, methanol, methylamine, acetate, etc. They metabolize only restricted range of substrates and are poorly characterized with respect to other metabolic, biochemical and molecular properties.

The sequence of the 16S ribosomal RNA (rRNA) from the archaebacterium Halobacterium volcanii has been determined by DNA sequencing methods. The archaebacterial rRNA is similar to its eubacterial counterpart in secondary structure. Although it is closer in sequence to the eubacterial 16S rRNA than to the eukaryotic 16S-like rRNA, the H. volcanii sequence also shows certain points of specific similarity to its eukaryotic counterpart. Since the H. volcanii sequence is closer to both the eubacterial and the eukaryotic sequences than these two are to one another, it follows that the archaebacterial sequence resembles their common ancestral sequence more closely than does either of the other two versions.

A rod-shaped Gram-positive methanogenic bacterium was isolated from marshy soil. It uses H2 – CO2 as sole energy and carbon source. Optimal growth occurs around 40 °C. The cell wall contains pseudomurein. The G + C content of the DNA is about 32 mol%. The DNA – DNA homology values of the isolate with Methanobacterium bryantii strains M.o.H. and M.o.H.-G. are 53 and 7%, respectively. The isolate is considered to be a new species within the genus Methanobacterium and it was named Methanobacterium uliginosum.
