Myeloperoxidase Immunohistochemistry as a Measure of Disease Activity in Ulcerative Colitis: Association With Ulcerative Colitis-Colorectal Cancer, Tumor Necrosis Factor Polymorphism and RUNX3 Methylation

Abstract: HDA is less sensitive than IHC and may underestimate inflammatory cell populations associated with UC-CRC. The epigenetic/genetic associations related to elevated MPO staining in UC-CRC may offer new methods for risk stratification and adjunctive screening tools.

“…30 Myeloperoxidase also demonstrates some promise as a marker of response to therapy. 63,64 Regarding mucosal healing, this evolving definition may ultimately include evaluation of both endoscopic and histologic remission, although standardized definitions of histologic response and remission are necessary and will require validation.…”

“…More recently, a cohort study by Gupta et al reported a significant link between histological inflammation and progression towards high-grade dysplasia or CAC, with a RR of 2 (Gupta et al, 2007). Finally, Garrity-Park et al showed an association between myeloperoxydase immunochemistry, TNF-α polymorphism and RUNX3 methylation and CAC (Garrity-Park et al, 2011). In addition to these clinical data, experimental results contribute to clarify the underlying cellular or molecular mechanisms explaining the link between inflammation and colorectal carcinogenesis in UC patients.…”

“…Based on the literature, the markers CD68 and CD163 were used to mark macrophages, myeloperoxidase (MPO) for neutrophils, and CD161 as well as CD244 for NK cells (34)(35)(36)(37). Slides were deparaffinized, hydrated in water and individual antigens stained using a DAKO Autostainer Plus.…”
