Mycobacterium tuberculosis CwsA Interacts with CrgA and Wag31, and the CrgA-CwsA Complex Is Involved in Peptidoglycan Synthesis and Cell Shape Determination

Abstract: cBacterial cell division and cell wall synthesis are highly coordinated processes involving multiple proteins. Here, we show that Rv0008c, a novel small membrane protein from Mycobacterium tuberculosis, localizes to the poles and on membranes and shows an overall punctate localization throughout the cell. Furthermore, Rv0008c interacts with two proteins, CrgA and Wag31, implicated in peptidoglycan (PG) synthesis in mycobacteria. Deletion of the Rv0008c homolog in M. smegmatis, MSMEG_0023, caused bulged cell po… Show more

“…This sequence has a plethora of surfaces for protein interactions to the extent that large hydrophobic residues that provide the majority of the van der Waals interactions for helix-helix binding are scarce. A cwsA and crgA double-deletion mutant showed that these proteins are required for cell wall PG synthesis and cell morphology (5). Fig.…”

“…Moreover, CrgA appears to have an important role in peptidoglycan (PG) formation during cell division, by recruiting PG synthases to the divisome (4). Reduced production of CrgA results in elongated cells and reduced growth rate (1), and loss of CrgA impairs PG synthesis (5). In addition to CwsA, the Mtb divisome involves other atypical players such as FipA (FhaB), ChiZ, and MtrB (6-8), and thus there is much yet to be learned about the participants in mycobacterial cell division (9).…”

“…In addition to FtsZ and the MtrB proteins, previous studies have shown that FtsI, FtsW, FipA, Wag31, CrgA, CwsA, RipA, and penicillin-binding protein A (PBPA) are other necessary septasomal components for the cell division in mycobacteria (6,(8)(9)(10)(11)(12)(13)(14)(15)(16). These studies also established pairwise interactions between FtsI and Wag31, FtsI and FtsW, FtsI and CrgA, CwsA and Wag31, and CrgA and PBPA.…”
