Myb -related genes of the Kyoho grape ( Vitis labruscana ) regulate anthocyanin biosynthesis

Abstract: Partial cDNAs of myb-related regulatory genes were isolated from the tetraploid Kyoho grape ( Vitis labruscana: V. labrusca x V. vinifera) and the expression patterns of the corresponding genes were studied. Since MybA gene expression is closely related to coloring and/or ripening of the berry (expression increases strongly with the commencement of coloring and berry softening, and is detected only in berry skin and flesh), full-length cDNAs for the gene were isolated from a mature-berry cDNA library. Three di… Show more

“…In grapes, VvMYBA1 is widely considered to be involved in the regulation of VvUFGT [56]. VvMYBA1 was down-regulated fivefold in sunlight-excluded ‘Jingxiu’ berry skin (and undetectable with qRT-PCR), which is consistent with the finding for ‘Cabernet Sauvignon’ grapes [11].…”

“…UDP-glucose:flavonol-3-O-glucosyltransferasae (UFGT, SSP:7411, AF000371) mRNA was also expressed specifically in the skin [51]. This enzyme plays a critical role in anthocyanin accumulation as retrotransposon-induced mutation of the MYB transcription factor (VvmybA1) gene [52] leading to its expression impairs anthocyanin accumulation in red grape varieties [53] with this mutation being common to many white grape varieties [5]. The enzymatic activity of recombinant UFGT has been documented in vitro [54].…”

“…In grapevine, most of the structural genes encoding the enzymes of the anthocyanins biosynthetic pathway have been identified (Sparvoli et al 1994;Boss et al 1996). Two adjacent transcription factors isolated from V. vinifera, VvmybA1 and VvmybA2, are involved in the control of the anthocyanins pathway (Kobayashi et al 2002;Kobayashi et al 2004;Walker et al 2007;Azuma et al 2008). The presence or absence of anthocyanins in grape berries segregates as a monogenic trait determined by a locus in linkage group 2 (Doligez et al 2002;Fischer et al 2004;Salmaso et al 2008).…”
