Mutagenicity and genotoxicity studies of aspartame

Abstract: Two studies were conducted to further assess its mutagenic and genotoxic potential. In a bacterial reverse mutation pre-incubation study, Salmonella typhimurium strains TA100, TA1535, TA98, and TA1537 and Escherichia coli WP2 uvrA were treated with aspartame at concentrations of up to 5000 μg/plate with or without metabolic activation and showed no mutagenic potential. Similarly, in vivo micronucleus testing of aspartame following gavage administration (500-2000 mg/kg body weight) to Crlj:CD1(ICR) strain SPF m… Show more

“…Although the safety of use of aspartame has been assessed by the Joint FAO/WHO Expert Committee on Food Additives (JECFA), the US Food and Drug Administration (FDA) and the European Food Safety Authority (EFSA), among others, few studies have tested its genotoxic properties in vivo and in vitro [67][68][69][70][71][72][73][74]. Unfortunately, none of these studies were conducted according to GLP [74,75]. Despite the lack of pharmaceutical studies in the GMP system, publications state that aspartame is not genotoxic [68,71,72,75,76].…”

In addition, research techniques have developed over the years and current methods are more accurate and reliable [75].…”

“…It was evaluated by Ames assay and in vivo micronucleus assay. As a result, no genotoxic or mutagenic potential was observed in either test (Otabe et al, 2019). The genotoxic effects of ascorbic acid, benzoic acid, citric acid and sorbic acid in human peripheral blood lymphocytes were investigated using in vitro micronucleus assay.…”

“…En el ensayo de variaciones en las frecuencias de células micronucleadas (MNs) para evaluar el potencial genotóxico, donde se comparan las frecuencias de micronúcleos en las muestras tratadas con las testigo y se evalúan su significancia. También en las muestras obtenidas se evalúa el efecto citotóxico mediante las comparaciones de los ratio entre los eritrocitos jóvenes y adultos observadas o por el porcentaje de eritrocitos jóvenes en el total de eritrocitos [18,19].…”
