Infection with hepatitis B virus (HBV) remains a worldwide health problem, and DNA-based vaccines against HBV have been tested for therapeutic applications. HBV possesses three envelope lipoproteins that are translated from a single reading-frame: large, middle, and small HBV surface antigens. Among these envelope proteins, the middle HBV surface antigen (MHBs) contains a constitutive N-linked glycosylation site at position 4 (Asn4) in the amino-terminal portion (MQWNSTTFHQ) of pre-S2 domain. Asn4 (shown in bold) is essential for secretion of viral particles and conserved among all serotypes of HBV, but its influence on the immunogenicity of MHBs remains unknown. Here, we constructed four MHBs genes carrying mutations, underlined, in the amino-terminal portion of pre-S2 domain. One mutant protein contains Q at position 4 (MQWQSTTFHQ). In addition, each of three mutant MHBs proteins contains a N-linked glycosylation site (N-X-S/T), relocated to position 5 (MQWQNTTFHQ), 6 (MQWQSNTSHQ) or 7 (MQWQSTNFTQ) in pre-S2 domain. The expression and immunogenic properties of mutant DNA vaccines were examined in 293T human renal epithelial cells and in BALB/c mice, respectively. We showed that Asn4 was critical for secretion and immunogenicity of MHBs. Moreover, the MHBs protein that carries a N-linked glycosylation site at position 5 or 7 retained the properties similar to wild-type MHBs. In contrast, the secretion-defective mutant protein carrying Asn at position 6 induced only marginal humoral and cellular immune responses in mice, despite the N-linked glycosylation. In conclusion, N-linked glycosylation at an appropriate position in pre-S2 domain is an essential requirement for DNA vaccine expressing MHBs.

RESUMENEsta revisión busca identificar las características de las lectinas vegetales con dominio de leguminosa que determinan su actividad insectistática e insecticida. Se realizó una búsqueda sistemática de literatura en las bases de datos Medline, Science Direct (Elsevier), Pubmed, Lilacs, SCOPUS, Web of Science (ISI) con el uso de palabras claves relevantes. La información recolectada fue clasificada y seleccionada en términos de su calidad y relevancia. La revisión tomó como base la información de las lectinas de Phaseolus vulgaris (PHA), Glechoma hederacea (Gleheda), Canavalia ensiformis (Concanavalina A), Griffonia simplicifolia (GSII) y Pisum sativum (PSA) y lectinas de leguminosas estudiadas en Colombia por el Grupo de Investigación en Proteínas. Se evidenció que las lectinas vegetales con dominio de leguminosa poseen rasgos estructurales caracterizados por un alto porcentaje de láminas beta asociados en estructuras diméricas o tetraméricas y que presentan varios sitios de reconocimiento a azúcares específicos, entre los cuales se destaca la manosa. Adicional a estas características, estas lectinas pueden interactuar con el sistema digestivo de insectos plaga y generar una disminución en su capacidad de absorción intestinal. Como conclusión, se puede señalar que es insuficiente la información sobre las señales que se activan y los efectos deletéreos de las lectinas con dominio de leguminosa sobre los insectos.Palabras clave: insectistático, insecticida, leguminosa, lectina. ABSTRACTThis review aims to identify the characteristics of legume lectins that determine their insecticidal and insectistatic activities. A systematic review of the literature was conducted in Medline, Science Direct (Elsevier), Pubmed, SCOPUS, Web of Science (ISI) and Lilacs databases by using relevant keywords. The information collected was classified and selected in terms of quality and relevance. Abstract:In the recent years, mushrooms are distinguished as important natural resources of immunotherapy which can be used as immunomodulating and immunostimulating in the management of some immunodeficiency diseases such as cancer, tumour, HIV, tuberculosis etc. Mushroom of the genus Pleurotus are good sources of several bioactive compounds which are able to augment or complement a desired immune response. Such bioactive compounds are polysaccharopeptides, polysaccharide-proteins, functional proteins (ubiquinone-9, nebrodeolysin, ubiquitin-like peptide and glycoprotein), glucans, proteoglycans and many others. Most of these bioactive compounds follow the immunomodulatory pathway mechanism of polysaccharide (β-glucan) from mushrooms by stimulating activities for both innate and adaptive immune systems. They proliferate and activate innate immune system components such as natural killer (NK) cells, neutrophils, and macrophages, and stimulate cytokines expression and secretion. These cytokines in turn activate adaptive immunity through the promotion of Bcells for antibodies production and stimulation of T-cell differentiation to T helper (Th1 and Th2) cells, which mediate cell and humoral immunities, respectively. In this review, the immunotherapeutic potential of oyster mushroom in relation to bioactive compounds produced is shown and this suggests that the oyster mushrooms are one of the most important natural products and functional foods.
