Muscarinic Receptor Activation of AMP-activated Protein Kinase Inhibits Orexigenic Neuropeptide mRNA Expression

Abstract: AMP-activated protein kinase (AMPK) plays a crucial role in both cellular and whole body energy homeostasis. Here we demonstrate that the muscarinic receptor agonist carbachol activates AMPK␣1-containing complexes in the human SH-SY5Y cell line via a mechanism specific for the AMPK upstream kinase, Ca 2؉ /calmodulin-dependent protein kinase kinase ␤. Activation of AMPK inhibits mRNA expression of the orexigenic neuropeptides Agouti-related peptide and melanin-concentrating hormone but surprisingly has no effec… Show more

“…As our results implicated a CaMKKβ‐AMPKα1‐dependent pathway, we used our previously characterized system of ionomycin (activator)/STO609 (inhibitor) treatment to modulate AMPK activity in a CaMKKβ‐dependent manner (Thornton et al . ). We exposed neurons to OGD in the presence of ionomycin or STO609 to determine the effect of extreme activation or inhibition of AMPK on cell survival.…”

“…AMPK is also activated by an increase in intracellular calcium ions, which is mediated by CAMKK2 (REFS [34][35][36]. Activation by calcium appears to play an important role in regulating AMPK in some tissues in response to hormones, such as VEGF-induced activation of muscarinic receptors in endothelial cells [66][67][68] or ghrelin acting on the ghrelin receptor in neuronal cells 66,69 . An exciting recent development is the finding that fructose 1,6-bisphosphate (FBP), a glycolytic intermediate, mediates glucose sensing by AMPK 70 .…”

“…CaMKKα and CaMKKβ [16] were originally discovered as CaMKs that acted upstream of CaMKI and CaMKIV. The Ca 2+ →CaMKK→AMPK pathway is triggered by a rise in cytosolic Ca 2+ without any requirement for an increase in AMP, and is responsible for AMPK activation in response to K + -induced depolarization in neurons [13], muscarinic activation in neuroblastoma cells [17], thrombin activation of endothelial cells [18], treatment of smooth muscle cells with vasoconstrictors [19] and stimulation of antigen receptors in T-cells [20]. …”
