Muscadine grape skin extract reverts snail-mediated epithelial mesenchymal transition via superoxide species in human prostate cancer cells

Abstract: BackgroundSnail transcription factor can induce epithelial-mesenchymal transition (EMT), associated with decreased cell adhesion-associated molecules like E-cadherin, increased mesenchymal markers like vimentin, leading to increased motility, invasion and metastasis. Muscadine grape skin extract (MSKE) has been shown to inhibit prostate cancer cell growth and induce apoptosis without affecting normal prostate epithelial cells. We investigated novel molecular mechanisms by which Snail promotes EMT in prostate c… Show more

“…The culture media was replaced with phosphate buffered saline containing 10μM DHE [32] and then cells were exposed to 30 min hypoxia/45 min reoxygenation. Fluorescence intensity was immediately quantified at the end of the reoxygenation by high performance multimode plate reader (Biotek, Winooski, VT).…”

“…Many literatures about the anti‐cancer effects of grape extracts were published, but only a few covered muscadine grapes extracts. Other phenolic components except anthocyanins in muscadine grape skin were totally ignored (Hudson and others ; Burton and others ). Some researchers separated different fractions and evaluated their anti‐cancer function (Yi and others ; Mertens‐Talcott and others ), but the fractions was only from the grape skin extracts.…”

“…Increasing evidence suggests that some Chinese herbal medicine, containing some bioactive compounds or a variety of phytochemicals, may inhibit EMT and reduce metastasis [ 5 7 ]. Thus, some herbs can be utilized as complementary and alternative treatments and as adjuvant therapy for conventional cytotoxic therapies.…”
