Musashi1 Promotes Non-Small Cell Lung Carcinoma Malignancy and Chemoresistance via Activating the Akt Signaling Pathway

Abstract: Background/Aims: Lung cancer is one of the leading causes for cancer mortality. The poor therapeutic outcome of non-small cell lung carcinoma (NSCLC) is mainly due to late diagnosis and chemoresistance. In this study, we investigated the role of Musashi1 (MSI1) in NSCLC malignancy and chemoresistance. Methods: Colony formation, MTT, glucose uptake and lactate production assays were employed to study lung cancer cell malignancy and chemoresistance. RT-PCR and Western blotting were performed to detect mRNA and p… Show more

“…By ectopic expression of MSI1 in NSCLC cell lines, A549 and H522, phosphorylated Akt is increased and cancer cell proliferation is elevated. After inhibition of the Akt pathway through treating cell lines with MK2206 small molecules, proliferation of cancer cells decreases [30]. MSI1 is also required for tumorigenesis in medulloblastoma and reduction of its expression decreases neurosphere formation, cell proliferation, and tumor growth.…”

“…However, mice with kinase-inactive mutants are not [43]. Since the p-AKT/survivin pathway plays an important role in drug resistance in NSCLC, targeting EGFR itself, instead of its tyrosine kinase activity, could be a more efficient way to treat NSCLC, especially when the cancer cells are resistant to TKIs [44,45].…”

“…NSCLC is typically carries a poor prognosis, with 5-year overall survival at around 15% [2]. Usually originating from peripheral lung tissues, lung adenocarcinoma (LAC) comprises more than 40% of lung cancers and has a high prevalence of distant metastasis [3]. The established curative method for managing LAC is surgical treatment.…”
