Murine thymomas induced by fractionated-X-irradiation have specific T-cell receptor rearrangements and characteristics associated with day-15 to -16 fetal thymocytes.

Abstract: We report here that specific T-cell receptor rearrangements were observed in fractionated-X-irradiation-induced murine leukemias. Consistent gamma-chain rearrangements, limited beta-chain rearrangements, and no detectable alpha-chain rearrangements were observed. Gene expression studies revealed that, in comparison with normal thymus tissue, expression of alpha T-cell receptor genes was lower in the thymomas, beta expression was much higher but approximately equal to that of normal thymocytes, and gamma expres… Show more

“…Cells. In vitro-adapted tumors (radiation leukemia virusinduced tumors and X-irradiation-induced tumors) of various inbred mouse strains were maintained in Dulbecco's modified Eagle's medium supplemented with 10% fetal bovine serum (FBS) (2,19,20 prepared from quiescent and activated splenocytes (A and C) and thymocytes (B) harvested at the indicated times following activation with the Tand B-cell mitogens, ConA (10 p.g/ml) and LPS (50 p.g/ml). After electrophoresis and Northern transfer, ERR transcripts (approximately 9 and 8 kb) were detected by hybridization to 32P-labelled ERR cDNA (BamHI-EcoRI) probe.…”

“…Some reports have shown that nearly all phenotypes of lymphoma cells corresponding to each stage of normal differentiation were induced by radiation (Gremers et al 1986) and carcinogens (Diamond et al 1987). Other reports have suggested that the lymphomas could result from the expansion of lymphoma cells arrested at a specific maturation stage (Amari and Meruelo 1987). These results suggests that is a strain difference in the predominant phenotype of lymphoma.…”

“…In the normal adult thymus less than 3% of the cells in the cortex express MEL-14 hi or IL-2R + [127,128] and the presence of MEL-14 hi may signify appearance of a leukemic clone [129]. Additionally, most of the TL tumors bear T-lymphocyte specific antigens Thy-1, Lyt-1, and Lyt-2 [130,131].…”
