Murine natural anti‐tumor antibodies. II. The contribution of natural antibodies to tumor surveillance

Abstract: The objective of this study was to investigate whether natural anti-tumor antibodies (NAb) contributed to the surveillance of small inocula of syngeneic tumors. Experiments were designed to distinguish between NAb and NK-mediated host resistance. Four approaches were used: (1) the isolation of tumor variants differing in their tumorigenicity and susceptibility to these mechanisms, and to activated macrophages; (2) a comparison of the effects of adjuvants on the modification of both host resistance and the acti… Show more

“…Macrophage-resistant tumor cell lines are rare (17,119,(129)(130)(131)(132), but Berd and Mastrangelo (133) reported the isolation of an LI210 line which was resistant to killing by Coo'nebacterium parvum (CP)-activated peritoneal exudate cells (PECs). Evidence that the effector PECs were macrophages included adherence, phagocytic activity, sensitivity to silica poisoning, absence of cytolytic activity in splenocytes from CP-treated mice and equivalent activity of PECs from both young and old CPtreated mice.…”

“…Variants differing in susceptibility to C'-dependent NAb lysis have been isolated by cloning from L5178Y lymphoma (135) and SL2 lymphomas (129). NAb lysis may involve synergistic cooperation between antibodies against multiple specificities on a tumor cell.…”

“…The new information obtained is that neither T cell nor NK activity was implicated in early CHP-100 cell death, which was seen in all mice within 24 hours. This early cell death could reflect other forms of natural immunity [32][33][34]. Alternatively, it might simply reflect inability of the murine host to support growth of the human cell line, perhaps coupled to the action of general inflammatory mechanisms.…”

“…The measurement of lysis by splenic NK cells from polyinosinic-polycytidylic-acid-stimulated syngeneic C3H/HeN and allogeneic CBA/J mice (100 pg i.p. 20 hr previously) was carried out in an 18-hr assay using "Cr-labelled target cells at effector-to-target ratios of 150:1, 75:l and 37.5:1 (Chow et al, 1981). Cytolysis was calculated according to the equation: Western blotting Expression of ras p21 was determined by Western blotting as described previously (Tough and Chow, 1991).…”
