Multiple Variables at the Leukocyte Cell Surface Impact Fc γ Receptor-Dependent Mechanisms

Abstract: Fc γ receptors (FcγR) expressed on the surface of human leukocytes bind clusters of immunoglobulin G (IgG) to induce a variety of responses. Many therapeutic antibodies and vaccine-elicited antibodies prevent or treat infectious diseases, cancers and autoimmune disorders by binding FcγRs, thus there is a need to fully define the variables that impact antibody-induced mechanisms to properly evaluate candidate therapies and design new intervention strategies. A multitude of factors influence the IgG-FcγR interac… Show more

“…This is accomplished through ligation with Fc gamma receptors (FcγRs), which forms a conduit of communication between the target cell (TC) and immune effector cell (1). The FcγRs are an assortment of transmembrane receptors expressed to varying levels on primarily innate, but also some adaptive, immune cells (2).…”

“…Many other variables, including antigen shape, size, and density as well as lipid environment and mobility, can also affect Fc presentation. Further, all these variables can change with antibody isotype, subclass and glycosylation as well as FcγR isotype, cellular subclass, FcγR expression and diversity as well as FcγR glycosylation and alleles (2).…”

“…The involvement of B-cell responses to Leishmania appears to be complex, and whether they participate in parasite persistence or killing remains unclear. Only four unique biological pathways were positively enriched in the NUCL group, including "Initiation of Nuclear Envelope Reformation", "Nuclear Envelope Reassembly" during mitotic cell cycle" 45 , "apoptosis induction by P53" 46 "and actin dynamic during phagocytosis" 47 .…”

“…Results leave the possibility of a role for FcgRIIIa. This point is relevant because FcgRIIIa, like FcgRI, is a classic activating receptor associating with an ITAM-signaling system; it might mediate a number of events previously attributed to FcgRIIIb (39). Alternatively, the balance in expression between the two isoforms may influence cellular responsiveness (39).…”
