Multiple Unit Pellet System (Mups Technology) for Development of Modified Release Fast Disintegrating Tablets: A Review

Abstract: Oral drug delivery system becomes challenging when the drug product needs to be delivered in modified release pattern in elderly patients, especially since it is difficult to swallow for them. Multiparticulates are the choice of dosage form when fast disintegration is desirable without loss of original release profile. Compressed multi-particulate system prepared by using pellets have several pharmacokinetic, pharmacodynamic, commercial and other advantages as mentioned henceforth in this review article. It in… Show more

“…Such drug-layered pellets (multiparticulates) can be placed in hard capsule shells or sachets and are intended to be sprinkled onto a teaspoon or tablespoon of soft food or liquid and swallowed immediately without chewing. The drug substance is contained in many independent units, which makes the formulation very flexible, so that the dose of the drug can be regulated only by the mass of the RSC multiparticulates applied [ 33 , 34 , 35 ].…”

“…Manufactured pellets demonstrate superior flow properties in comparison to their individual counterparts, which allows for better flow from the hopper to the tablet die during compression, thereby increasing content uniformity due to less mass variation [ 137 , 149 ]. Whilst better powder flow results in higher processing speeds, such compositions also demand lower lubricant concentrations, thereby lowering processing costs [ 144 , 150 ]. This technique is well suited to formulations where differences in particle size of the APIs and excipients are a concern, as pellets negate concerns of segregation within a mixture.…”

“…Incompatible drugs can be combined into one preparation form and can be designed to produce different release profiles from the same drug or from different drugs. MUPS is made from pellets coated with a reservoir with a characteristic of polymer coating that can withstand compression strength, in which the coating must not be crushed or even change shape 19 .…”
