Multiple dose-dependent roles for Sox2 in the patterning and differentiation of anterior foregut endoderm

Abstract: Sox2 is expressed in developing foregut endoderm, with highest levels in the future esophagus and anterior stomach. By contrast, Nkx2.1 (Titf1) is expressed ventrally, in the future trachea. In humans, heterozygosity for SOX2 is associated with anopthalmiaesophageal-genital syndrome (OMIM 600992), a condition including esophageal atresia (EA) and tracheoesophageal fistula (TEF) luminal mucus-producing cells, fewer p63 + cells, and ectopic expression of genes normally expressed in glandular stomach and intestin… Show more

“…Indeed, mutants showed severe defects of stomach, lung, and eye development, confirming the role of Sox2 in the development of foregut endoderm and retina [33][34][35][36]. Sox2 loxP/lacZ ;Blimp1 Cre mutant eyes showed a strong reduction of the lens vesicle at 12.5 dpc, while at later stages (17.5 dpc), also the retina was severely impaired (Supporting Information Fig.…”

“…In the developing lung, Sox2 is normally expressed in endoderm progenitors of the proximal airways and is absent in the distal endoderm (Fig. 3A) (Que et al, 2007). Conversely, Sox9 is expressed at the highest levels in the distal lung endoderm progenitors and expression in distal progenitors decreases significantly so that by E16.5, Sox9 is detected in only a small subset of cells in the terminal distal endoderm (Fig.…”

“…mesodermal derivatives, but how these interact with endoderm-autonomous regional cuing needs much more detailed study. There are several notable endodermal region-or organ-specific transcription factors (TFs) that are differentially expressed along the A-P axis, which include Six1 and Six4 (pharyngeal endoderm) (Zou et al, 2006), Sox2 (stomach) (Que et al, 2007), Nkx2.1 (lung) (Minoo et al, 1999), Hhex (liver) (MartinezBarbera et al, 2000;Keng et al, 2000), Pdx1 (pancreas, duodenum, posterior stomach, common bile duct) (Offield et al, 1996), and the caudal homeobox genes Cdx1 and Cdx2 (intestine) (Beck et al, 1995(Beck et al, , 2003.…”
