Multiple catalytically active thioredoxin folds: a winning strategy for many functions

Abstract: The Thioredoxin (Trx) fold is a versatile protein scaffold consisting of a four-stranded β-sheet surrounded by three α-helices. Various insertions are possible on this structural theme originating different proteins, which show a variety of functions and specificities. During evolution, the assembly of different Trx fold domains has been used many times to build new multi-domain proteins able to perform a large number of catalytic functions. To clarify the interaction mode of the different Trx domains within a… Show more

“…Pedone et al have proposed that thermophilic protein disulfide oxidoreductase (PDO), which is a member of the thioredoxin family, plays a main role in the cytoplasmic disulfide formation in thermophilic microorganisms, such as S. solfataricus (9,10). The microorganisms possessing thermophilic PDO seem to agree with those listed by Yeates's group as the producers of abundant disul-fide-bonded proteins in the cytosol (6,7).…”

“…3C) and that they indeed feature the two characteristics of the Prx family. An additional helix in the variable Trx-fold insertion, αD in the Dio-insertion, is present in Prx but also in some other Trx-fold protein families (6). The N-terminal βN1/2-Θ1 module found in Dio3 cat (Fig.…”

“…The deiodinase crystal structure now suggests an answer to this question. The Trx superfamily comprises protein families with different functions, characterized by family-specific modifications to the Trx fold (6).…”

“…These proteins are generally members of the thioredoxin superfamily and include proteins such as the thioredoxins, glutaredoxins, peroxiredoxins and protein disulfi de isomerases (PDIs) [ 5 ] and also proteins associated with the function of these redox proteins [ 6 ]. Human thioredoxin-1 (Trx-1) is part of a system of proteins which include a truncated form of Trx termed Trx80, thioredoxin reductase (TrxR) and a natural inhibitory protein known as thioredoxin-interacting protein (TXNIP).…”
