Multiplatform untargeted metabolomics

Abstract: Metabolomics samples like human urine or serum contain upwards of a few thousand metabolites, but individual analytical techniques can only characterize a few hundred metabolites at best. The uncertainty in metabolite identification commonly encountered in untargeted metabolomics adds to this low coverage problem. A multiplatform (multiple analytical techniques) approach can improve upon the number of metabolites reliably detected and correctly assigned. This can be further improved by applying synergistic sam… Show more

“…Multivariate analysis (principal component analysis, PCA) was firstly conducted to reflect the general differences in metabolites among each group. 25 Figure 4a shows that the samples of the control and model groups were clustered separately in the PCA score plot, and a clear separation in the dispersion area was observed, indicating that the lung metabolic profile of model group mice was changed. Similarly, the samples of the model and HPM groups were also clustered separately in the PCA score plot with a clear separation in the dispersion area, indicating that the lung metabolic profiles of the mice in the HPM group were changed.…”

“…This multiplatform metabolomics study enables us to discover and quantify a wide range of distinct urine metabolites or metabolite species, boosting metabolome coverage. Numerous research groups also combine two or more analytical approaches to assist enhance metabolite coverage [ 74 , 81 ].…”

“…These advances have led to the application of multiomics approaches to defining trajectories associated with T2D by integrating the impact of silent microvascular damage through cohorts of patients using high-throughput longitudinal phenotyping [9]. Metabolomics is a novel high-throughput profiling technique that can comprehensively reveal the levels of small-molecule metabolites of size <1500 Da [10]. Because the metabolome is downstream from the genome, transcriptome, and proteome, it represents a more sensitive level of organization for understanding complex biological systems, and can provide a highly integrated profile of biological status [9,11].…”
