Multilocus Sequence Typing of Klebsiella pneumoniae Nosocomial Isolates

Abstract: A multilocus sequence typing (MLST) scheme was developed for Klebsiella pneumoniae. Sequences of seven housekeeping genes were obtained for 67 K. pneumoniae strains, including 19 ceftazidime-and ciprofloxacinresistant isolates. Forty distinct allelic profiles were identified. MLST data were validated against ribotyping and showed high (96%) discriminatory power. The MLST approach provides unambiguous data useful for the epidemiology of K. pneumoniae isolates.

“…The khe gene was amplified using the following primers: forward (5 0 -TGATTGCATTCGCCACTGG-3 0 ) and reverse (5 0 -GGTCAACCCAACGATCCTG-3 0 ) (He et al 2012). The rpoB gene was amplified using the primers VIC3 (5 0 -GGCGAAATGGCWGAGAACCA-3 0 ) and VIC2 (5 0 -GAGTCTTCGAAGTTGTAACC-3 0 ) (Diancourt et al 2005). Each of 50 lL PCR reactions contained 25 lL 29Taq Master Mix (SinoBio, Shanghai, China), 0.4 lM of each primer, and 2 lL genomic DNA.…”

“…Housekeeping genes are very useful in developing multilocus sequence typing (MLST) scheme for discriminating K. pneumoniae, which further provides unambiguous data useful for the epidemiology of K. pneumoniae. For instance, sequences of seven housekeeping genes, i.e., rpoB, gapA, mdh, pgi, phoE, infB and tonB, have been successfully used to develop a MLST scheme for K. pneumoniae (Diancourt et al 2005). On the other hand, the identification of K. pneumoniae strains were generally confirmed in more simple ways, e.g., by rpoB and/or gyrA gene sequences (Brisse and Duijkeren 2005;Brisse and Verhoef 2001;Brisse et al 2004).…”

“…Additionally, four/eight K. pneumoniae isolates were grouped into the same cluster (Kp1) and two of these isolates also possessed the qnrB2 allele. According to MLST analysis (Diancourt et al 2005), seven/eight K. pneumoniae isolates were typed as ST11 (Table). …”
