Multifunctional Nanofibers for Specific Purification and Release of CTCs

Abstract: Recovering pure and viable circulating tumor cells (CTCs) from blood has been a challenging task for molecular characterization and functional analysis, which has attracted wide attention these days. Herein, we fabricate a thermoresponsive chitosan nanofiber substrate to effectively capture, purify, and release the target cancer cells, assisted by PNIPAAm brushes and DNA hybridization. The PNIPAAm brushes are designed to enable WBCs to detach from aptamer-PNIPAAm-chitosan-nanofiber (aptamer-P-CNFs) surfaces du… Show more

“…Moreover, the spacer-to-transferrin ratio played also an important role on the system efficiency. Three linkers per transferrin gave the highest capturing values for all the polymer lengths that reached a top value of 81% for PEG 8 . Moreover, the use of a bigger thermoresponsive polymer on the construction of the MNG had a negative effect on the capturing ability of the system.…”

“…The 1:1 ratio did not guarantee effective CTC capturing, giving values of about 30%. With a ratio of 1:3, about 60% of the CTCs were captured with PEG 4 and PEG 12 and, even more interestingly, an 81% capturing efficiency was achieved with PEG 8 . Above a ratio of 1:3, the increase of linker per Tf decreased the capturing efficiency of the system.…”

“…These brushes are designed to enable white blood cells to detach from aptamer‐poly( N ‐isopropylacrylamide)/chitosan nanofiber surfaces during the conformational transition. Meanwhile, these specific captured circulating tumor cells (CTCs) are retained at a high purity …”
