The avian embryos growing outside the natural eggshell (ex ovo) were observed since the early 19th century, and since then chick embryonic structures have revealed reaching an in-depth view of external and internal anatomy, enabling us to understand conserved vertebrate development. However, the internal environment within an eggshell (in ovo) would still be the ideal place to perform various experiments to understand the nature of avian development and to apply other biotechnology techniques. With the advent of genetic manipulation and cell culture techniques, avian embryonic parts were dissected for explant culture to eventually generate expandable cell lines (in vitro cell culture). The expansion of embryonic cells allowed us to unravel the transcriptional network for understanding pluripotency and differentiation mechanism in the embryos and in combination with stem cell technology facilitated the applications of avian culture to the next levels in transgenesis and wildlife conservation. In this review, we provide a panoramic view of the relationship among different cultivation platforms from in ovo studies to ex ovo as well as in vitro culture of cell lines with recent advances in the stem cell fields.

The major challenge in treating cancers with ATRA is the limited availability inside the cell and resistance developed in prolonged treatment. We made an attempt for co-treatment of human NSCLC cell lines (A549) with ATRA and its isomeric precursor (9cisRA). In this study, the growth inhibitory effect of ATRA, 9cisRA and combination of both were tested in A549 cells by MTT and Trypan blue assays. As the effects of retinoid are mediated through their receptors, their gene expression levels were analyzed by RT-PCR. The target gene receptor, RAR-β protein expression, was analyzed by immunocytochemistry. The cancer cell (A549) growth inhibitory effect was significantly (p ≤ 0.001) enhanced in combination treatment when compared with the result of individual treatments. The mRNA expression levels of both RAR-β and RXR-β were found to be increased in co-treatment (band density of 0.75 and 0.806, respectively) when compared with 9cisRA treatment (0.25 and 0.112) and ATRA treatment (0.01 and 0.081). A concomitant enhancement in the target RAR-β protein expression was observed in co-treated cells when compared with individual treatments. We thus conclude that the co-treatment had increased the availability of ATRA, by isomerization of the 9cisRA which then resulted in an increased expression of both RAR-β and RXR-β receptors and the target protein RAR-β which in turn inhibited lung cancer cell growth. Our study results have explored the mechanism of synergistic effect of co-treatment with ATRA and 9cisRA and further preclinical studies are necessary to validate the application of co-treatment of retinoid in clinical use.

Evaluating the effects of calcium supplementation for chicken embryos cultured in a novel biomimetic transparent eggshell

The chicken embryo model has long been touted as an attractive alternative for research due to its high growth rate, ease of manipulation, and logistical advantages. There are two primary techniques associated with this model; in-ovo and ex-ovo culture. The in-ovo culture is a method that allows the observation of the embryo through a small window, but poses challenges such as poor visibility, limited embryonic accessibility, and lengthy preparation time.Conversely, the ex-ovo culture functions by cultivating the embryo outside the shell. This addresses some issues highlighted above, but presents new hurdles such as poor viability and sustainability due to a lack of a calcemic eggshell and an unnatural environment. As such, this study aims to optimize ex-ovo culturing for chicken embryos by introducing the usage of a novel biomimetic, transparent polydimethylsiloxane eggshell coupled with calcium supplementation. This polydimethylsiloxane system enables a high degree of optical clarity in a three-dimensional state and retains an elliptical geometry of an egg. Benefits of utilizing such an approach can include enhanced visibility, improved accessibility, as well as overall design robustness. Alongside utilizing eggshell powder as a calcium supplement, the amount and timing of this addition were also investigated to assess optimization strategies. As a result, it was discovered that applying eggshell powder during the earlier embryonic stages was more beneficial in terms of overall growth and viability. Enhanced viability rates were also observed for all groups supplemented with eggshell powder. Additionally, a lower calcium quantity usage was perceived to generate more favorable responses in certain key characteristic aspects.These findings suggest that an extraembryonic calcium supply regulation may be time and quantity-dependent. On this basis, the concept of timing and quantity-based calcium supplementation for chicken ex-ovo models should be considered when designing experimental goals.
