Multicolor Spectral Karyotyping of Human Chromosomes

Abstract: The simultaneous and unequivocal discernment of all human chromosomes in different colors would be of significant clinical and biologic importance. Whole-genome scanning by spectral karyotyping allowed instantaneous visualization of defined emission spectra for each human chromosome after fluorescence in situ hybridization. By means of computer separation (classification) of spectra, spectrally overlapping chromosome-specific DNA probes could be resolved, and all human chromosomes were simultaneously identifie… Show more

“…The chromosome spreads were subjected to spectral karyotyping (SKY), a recently developed molecular cytogenetic technique in which each chromosome is painted a di erent color by hybridization to¯uorescent probes (Schrck et al, 1996). SKY allows an unambiguous identi®cation of each chromosome, an extremely di cult task in early embryos by G-or DAPIbanding.…”

“…Subsequently, spreads were hybridized with a probe mixture supplied by Applied Spectral Imaging (ASI, Carlsbad, Calif.). The probe mixture contained 24 whole chromosome painting probes, labeled with combinations of five different fluorescent dyes (Spectrum Green, Spectrum Orange, Texas Red, Cyanine [Cy] 5 and Cy5.5) [16]. The probe (SKY™ mixture) was denatured at 75°C for 7 min and incubated at 37°C for 1 hr.…”

“…This technique, along with the technically similar spectral karyotyping (SKY), has greatly facilitated the identification of chromosome material involved in chromosomal rearrangements even in sup-optimal chromosome preparations. 12 Used in combination, the 2 techniques allow chromosomal alterations in human tumours to be characterised more extensively than was previously possible by G-banding alone. A genome wide search for chromosomal abnormalities in SCLC will identify regions of deletion and gain likely to contain genes involved in the development of this disease.…”
