In regard to the knowledge of provoking a worldwide resistance against antibiotics due to incorrect application and the resulting uptake of residues via the food chain, the European Union set maximum residue levels (MRLs) for these substances in animal derived products to protect the customers. In respect of these MRLs a multi-class UHPLC-MS/MS multiclass method has been developed for the simultaneous determination of 30 substances from different compound groups (quinolones, macrolides, lincosamides, β-lactams, sulphonamides, diamino-pyrimidine derivates and tetracyclines) in various kinds of dairy products. Since this method should be suitable for routine laboratory use, sample preparation requires an easy and fast approach to ensure high sample throughput. Therefore a sample preparation with C18EC dSPE bulk sorbent as a combination of the easy and quick sample preparation provided by QuEChERS and the clean-up principle of reversed phase SPE was applied. This method was validated for all compounds in all matrices in compliance with the requirements of Commission Decision 2002/657/EC.

A new, simple and fast method was developed for the determination of multi-class antibiotic residues in honey (sulfonamides, tetracyclines, macrolides, lincosamides and aminoglycosides). Separation and determination were carried out by LC-MS/MS. During sample preparation, various parameters affecting extraction efficiency were examined, including the type of solvent, pH, efficiencies of cleavage of N-glycosidic linkages by hydrochloric acid, ultrasonic extraction and its duration compared with shaking, along with dispersive SPE clean-up. Experiments with fortified samples demonstrated that 10 min of ultrasonic treatment with acidified methanol (HCl 2 M) followed by dispersive SPE clean-up with 50 mg PSA gave an effective sample preparation method for several classes of antibiotics in honey. Anhydroerythromycin A, erythromycin A enol ether and desmycosin were used as markers for the presence of erythromycin A and tylosin A in honey samples. The method was validated according to European Commission Decision (EC) No. 2002/657. The recoveries of analytes ranged from 85% to 111%. Repeatability and intra-laboratory reproducibility were < 20.6% and 26.8%, respectively. Decision limit (CCα) and detection capability (CCβ) ranged from 6 to 9 µg kg and from 7 to 13 µg kg, respectively, except for streptomycin and neomycin, which showed slightly higher CCα at 25 µg kg and CCβ at 34 µg kg. Finally, the method was applied to the honey test material 02270 through a FAPAS proficiency test (PT) for the determination of tetracyclines. PT results were obtained within a z-score range of ±2, proving that the validated method is suitable for routine analysis to ensure the quality of honey.

An accurate, reliable and fast multianalyte/multiclass ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was developed and validated for the simultaneous analysis of 23 pharmaceuticals, belonging to different classes amphenicols, sulfonamides, tetracyclines, in honey samples. The method developed consists of ultrasonic extraction followed by UPLC-ESI-MS/MS with electrospray ionization in both positive mode and negative mode. The influence of the extraction solvents and mobile phase composition on the sensitivity of the method, and the optimum conditions for sample weight and extraction temperature in terms of analyte recovery were extensively studied. The identification of antibiotics is fulfilled by simultaneous use of chromatographic separation using an Acquity BEH C 18 (100 mm x 2.1 mm, 1.7 µm) analytical column with a gradient elution of mobile phases and tandem mass spectrometry with an electrospray ionization. Finally, the method developed was applied to the determination of target analytes in honey samples obtained from the local markets and several beekeepers in Muğla, Turkey. Ultrasonic-extraction of pharmaceuticals from honey samples is a well-established technique by UPLC-ESI-MS/MS, the uniqueness of this study lies in the simultaneous determination of a remarkable number of compounds belonging to 23 drug at the sub-nanogram per kilogram level.Keywords: antibiotics; fast method; honey; UPLC-ESI-MS/MS; chloramphenicol.Practical Application: Honey is one of the healthy foods across the world. Food safety is an important issue regarding residues of pharmaceutical drugs. The developed method proposed simultaneous analysis of analysis of twenty-three veterinary pharmaceuticals in honey. A fast and sensitive multianalyte/multiclass ultra-performance liquid chromatography-tandem mass spectrometry with electrospray ionization (UPLC-ESI-MS/MS) method provides the analysis of 50 honey samples in 50 minutes for food and laboratory sectors, and benefits safe food consumption.
