The Treatment of Endomorphous and Psychogenic Depressions with a Fixed Combination of Amitriptyline/Flupenthixol (Lu 7410)

“…Based on these findings we conclude that Lu 7410 has an activating effect in low doses (which are described at daytime), while in higher doses (which are given at bedtime), it is anxiolytic, tranquilizing and sleep-induc ing. Indeed, the improvement of sleep disturbances was the most pronounced somatic finding, which stands in sharp contrast to the reports about Deanxit®, which was found to frequently induce insomnia (22,24). Other psychosomatic complaints such as headaches, cardiac sensations, and sweating improved alsobut especially so in psychogenic depressed patients.…”

“…So described Strotzka (24), Cermak and Ringel (3), Ravn (19) and Simmelsgaard et al (22) a drug combination similar to the present investigated one -flupenthixol/melitracen (Deanxit®) as a good antidepressive drug and as a psycho-activator to be given in depressions and psychovegetativeneurotic disorders. Specifically they reported rapid improvement in efficacy, inhibition, depressed mood, passivity, exhaustion, despair and lack of initiative.…”

“…Reviewing 9 studies about the combination of perphen azine/amitriptyline, Freeman (7) found 45 % of psychoneurotic and depressed patients as markedly improved, which is, however, similar to the percentage one expects with amitriptyline or another thymoleptic drug alone. Simmelsgaard et al (22) investigated the clinical efficacy of Deanxit® as compared with flupenthixol in predominantly neurotic patients, and found the combination only slight ly (but not significantly) superior to flupenthixol alone. These contradictory findings could very well be due to the different biochemical pharmacokinetic interactions between the single components of a drug combination.…”
