Background Excessive activation of NLRP3 inflammasome and down-regulation of Sirt1/Smad3 deacetylation pathway play a significant role in the evolution of renal fibrosis. In China, it has been well known that Chinese herbal medicine is markedly effective in treating chronic kidney disease (CKD). Shen Shuai IIRecipe (SSR) has been used clinically for more than 20 years and has been confirmed to be effective in improvements of renal function and fibrosis. However, the specific mechanisms under the efficacy require further research. The purpose of this study was to evaluate whether SSR could alleviate renal injury and fibrosis by regulating NLRP3 inflammasome and Sirt1/Smad3 deacetylation pathway. Methods Four weeks after 5/6 ablation/infarction (A/I) surgery, Sprague-Dawley rats were randomly divided into the following groups: sham operation group, 5/6 (A/I) group, 5/6 (A/I) + SSR group, and 5/6 (A/I) + Losartan group (5/6 (A/I) + Los). After 8 weeks intervention,we mainly assessed the severity of renal injury and fibrosis along with the activation of NLRP3 inflammasome and Sirt1/Smad3 deacetylation pathway. Results SSR significantly attenuated renal injury and fibrosis in the remnant kidneys. In addition, we found that SSR effectively inhibited activation of NLRP3/ASC/Caspase-1/IL-1βcascade, decreased inflammatory infiltration and up-regulated Sirt1/Smad3 deacetylation pathway. Conclusions SSR could contribute to renal protection by inhibiting the activation of NLRP3 inflammasome and, furthermore, strengthen the antifibrotic effects by up-regulating Sirt1/Smad3 deacetylation pathway in 5/6 renal (A/I) model.

Breast cancer (BC) exhibit a high level of mortality rate and incidence but a comprehensive treatment is unavailable, which can successfully eradicate this fatal disease. Novel therapeutic strategies have been intensively studied to eradicate BC, including the use of traditional medicines and phytochemical (herbal compounds). These compounds have been studied in many aspects, such as targeting cell cycle and key tumor-associated metabolic pathways. In this study, we reviewed the literature for the potential therapeutic application of herbal compounds against breast cancer. Several terms, including "Herbal compounds", "Breast Cancer" and "Traditional medicines", were searched in PubMed and Web of Science databases. The search results were narrowed down using many filters and the resultant literature was critically analyzed. The enormous number of articles showed the promising potential of herbal compounds as anticancer therapy. The comprehensive evaluation of the literature suggested the anti-angiogenic, anti-proliferative, anticancer stem cells, anti-inflammatory/antioxidant, anti-metastatic, and epigenetic effects of herbal compounds as anticancer effects against BC. Despite this enormous data, there is lack of sufficient demonstration of the consumption of herbal compounds in a regular as anticancer strategy in clinical trials. Therefore, given its promising anticancer potential, focus should be shifted towards the regular use of herbal compounds in anticancer strategies.

Background Breast cancer is the leading cause of cancer-related death in women worldwide. Although traditional Chinese medicine (TCM) is commonly used by patients with breast cancer, little is known about TCM prescriptions for breast cancer. This study investigated the effects of a new TCM formula, T33, comprising Radix Kansui , Rheum rhabarbarum , Paeonia lactiflora , Jiangbanxia , and Zhigancao on breast cancer cells in vitro and in vivo. Methods To evaluate the effects of T33 on human breast cancer, HMEpiC, MDA-MB231 and MCF-7 cells were treated with different concentrations of T33 and then analyzed using MTT and Transwell migration assays. To elucidate the involvement of autophagy in the T33-induced death of MDA-MB231 and MCF-7 cells, immunofluorescence staining with LC3-II-specific antibodies was performed. Tumor xenografts were generated by subcutaneously injecting either MDA-MB231 or MCF-7 cells into BALB/c nude mice to determine the effects of T33 on these cell lines in vivo. Results The experimental results revealed that 0.1 mg/mL, 0.5 mg/mL, 2.5 mg/mL, 5 mg/mL and 10 mg/mL T33 significantly inhibited the proliferation and invasion of MDA-MB231 and MCF-7 cells. Moreover, significant autophagy was observed in MDA-MB231 and MCF-7 cells in the presence of 2.5 mg/mL, 5 mg/mL and 10 mg/mL T33. An animal study further revealed that both low (200 mg/kg) and high (600 mg/kg) doses of T33 inhibited the proliferation of xenografted breast cancer cells in BALB/c nude mice. Conclusion These findings demonstrate for the first time that T33 has potential in the treatment of breast cancer owing to its antiproliferative effects and induction of autophagy.
