Multi-responsive polypeptide hydrogels derived from N-carboxyanhydride terpolymerizations for delivery of nonsteroidal anti-inflammatory drugs

Abstract: A polypeptide-based hydrogel system, when prepared from a diblock polymer with a ternary copolypeptide as one block, exhibited thermo-, mechano- and enzyme-responsive properties, which enabled the encapsulation of naproxen (Npx) during the sol–gel transition and its release in the gel state. Statistical terpolymerizations of l-alanine (Ala), glycine (Gly) and l–isoleucine (Ile) NCAs at a 1:1:1 feed ratio initiated by monomethoxy monoamino-terminated poly(ethylene glycol) afforded a series of methoxy poly(ethyl… Show more

“…Recently, Fan et al . reported a peptide-based hydrogel system [ 62 ]. A polypeptide hydrogel system prepared from ternary sequences of l-alanine, glycine and l-isoleucine within a polypeptide segment of a PEG-peptide diblock structure exhibited thermo-, mechano- and enzyme-responsive properties, which enabled the encapsulation and release of naproxen (Npx) (Fig.…”

“…The synthesis of well‐defined polypeptide architectures via ROP of N ‐carboxyanhydride (NCA) has received increasing attention during the last decade . Several reports have been published depicting the synthesis of micelles, vesicles, and nanogels by controlled ROP of NCAs . Mechanism of ROP (normal amine mechanism, NAM) (Figure B), involves the attack of a primary amine (nucleophile) to initiate polymerization of NCA leading to the formation of a polypeptide, whose molecular weight can be controlled by regulating NCA to amine ratio.…”

“…Amongst all types of hydrogels, peptide-based hydrogels have self-assembly ability for hydrogel formation through hydrogen bonding, aromatic (cation-π, π-π), and hydrophobic/hydrophilic interactions [ 12 , 13 , 14 , 15 ]. Additionally, peptide segments can adopt secondary conformations (random coil, β-sheet/β-turn) inside their gel network to enhance the mechanical properties of hydrogels [ 16 , 17 , 18 , 19 , 20 ]. As a type of synthetic hydrogel, polypeptides can modify the characteristics of hydrogels by varying the functional groups on the side chain or the chain composition on the backbone [ 21 , 22 , 23 ].…”
