Muga (Antheraea assamensis) is an economically important silkmoth endemic to North-eastern part of India and is the producer of the strongest known commercial silk. However, there is a scarcity of -omics data for understanding the organism at a molecular level. Our present study decodes the complete mitochondrial genome (mitogenome) of A. assamensis and presents comparative analysis with other lepidopterans. Mitogenome is a 15,272 bp long AT rich (~80.2%) molecule containing 37 genes (13 PCGs, 22 tRNAs, 2 rRNAs) and a 328 bp long control region. The muga silkworm, Antheraea assamensis, is exclusively present in the northeastern regions of India and rearing of this silkworm is a vocation unique to this region in the world. Through culture-dependent techniques, generic identification using 16S ribosomal RNA probes, diversity analysis and qualitative screening for enzyme activities, our studies have identified a number of bacterial isolates, viz. Bacillus spp., Serratia marcescens, Stenotrophomonas maltophilia, Pseudomonas stutzeri, Acinetobacter sp. and Alcaligens sp., inhabiting the gut of the muga silkworm. Analysis of the culturable bacterial community from the gut of An. assamensis revealed that Bacillus (54%) was the predominant bacterial genus followed by Serratia (24%), Pseudomonas (10%) and Alcaligens (6%). Significant differences in the Shannon-Wiener (H') and Simpson (D) diversity indices of gut bacteria were recorded for An. assamensis collected from different regions. H' and D values were found to be highest for An. assamensis from the Titabar region (H' = 4.73 ± 0.43; D = 10.00 ± 0.11) and lowest for individuals from the Mendipathar region (H' = 2.1 ± 0.05; D = 0.04 ± 0.00) of northeastern India. Qualitative screening for enzyme activities identified about 26 gut bacterial isolates having significantly higher cellulose, amylase and lipase activities. These isolates probably contribute to the digestion and nutrition of their host insect, An. assamensis.

Abstract. Fast and accurate discrimination of pesticide residue in mulberry leaf is very important for sericulture and silk textile industry. Therefore, a hyperspectral imaging approach with the spectral range of 390-1050 nm was used for the discrimination of pesticide residue in mulberry leaf. 120 mulberry leaves samples including 60 samples without pesticide residue and 60 samples with pesticide residue were imaged by the VIS-NIR hyperspectral imaging system. ENVI software was used to explore the region of interest and extract the corresponding spectral data. Partial least square discriminant analysis (PLSDA) was used to establish the discriminative model for the discrimination of pesticide residue in mulberry leaf and the model achieved 98.33% calibration accuracy and 93.33% prediction accuracy. A total of 9 important wavelengths were selected according to the regression coefficient in the PLSDA model. A simplified PLSDA model was developed based on the important wavelengths and it achieved similar results (96.67% and 93.33%). The results showed that the model based on the selected important wavelengths was comparable to the model based on the full wavelengths, and it was feasible to use hyperspectral imaging technology for discrimination of pesticide residue in mulberry leaf.
