Oral Treatment of Spontaneously Hypertensive Rats with Captopril-Surface Functionalized Furosemide-Loaded Multi-Wall Lipid-Core Nanocapsules

Multi-wall lipid-core nanocapsule (MLNC) functionalized with captopril and nanoencapsulating furosemide within the core was developed as a liquid formulation for oral administration. The nanocapsules had mean particle size below 200 nm, showing unimodal and narrow size distributions with moderate dispersity (laser diffraction and dynamic light scattering). Zeta potential was inverted from −14.3 mV [LNC-Fur(0,5)] to +18.3 mV after chitosan coating. Transmission electron microscopy and atomic force microscopy showed spherical structures corroborating the nanometric diameter of the nanocapsules. Regarding the systolic pressure, on the first day, the formulations showed antihypertensive effect and a longer effect than the respective drug solutions. When both drugs were associated, the anti-hypertensive effect was prolonged. On the fifth day, a time effect reduction was observed for all treatments, except for the nanocapsule formulation containing both drugs [Capt(0.5)-Zn(25)-MLNC-Fur(0.45)]. For diastolic pressure, only Capt(0.5)-Zn(25)-MLNC-Fur(0.45) presented a significant difference (p < 0.05) on the first day. On the fifth day, both Capt(0.5)-MLNC-Fur(0.45) and Capt(0.5)-Zn(25)-MLNC-Fur(0.45) had an effect lasting up to 24 h. The analysis of early kidney damage marker showed a potential protection in renal function by Capt(0.5)-Zn(25)-MLNC-Fur(0.45). In conclusion, the formulation Capt(0.5)-Zn(25)-MLNC-Fur(0.45) proved to be suitable for hypertension treatment envisaging an important innovation.

We reported a novel transport mechanism of curcumin, independent of improved solubility, which involved direct contact of amorphous solid particles with the cell membrane. This mechanism has potential as a novel systemic delivery system of poorly water-soluble drugs. In this study, the transport mechanism of furosemide (FUR), which is transported by the same novel mechanism, was examined. In vitro cell permeation studies under air-interface conditions (AICs) revealed that the permeation from powders sprayed on cell monolayers was significantly higher than that under liquid-covered conditions (LCCs) from their solutions. The permeation from amorphous solid particles was faster than that from crystals. Similar results were derived from in vitro studies using an artificial membrane, with which the permeation of FUR could be examined without water. These findings clearly indicated that the transport mechanism of FUR is the same as that of curcumin. For the application of this new transport mechanism, the in vivo absorption of FUR was examined after pulmonary insufflation, which allows the solid particles to make direct contact with the epithelial cells. Pulmonary absorption of FUR from the amorphous powder was almost complete and was faster than that after intragastric administration of the solution, suggesting that FUR was absorbed from the lung by the same mechanism as the in vitro study. This new transport mechanism, which is independent of water dissolution, could be exploited to develop a novel delivery system for poorly water-soluble drugs, using pulmonary powder inhalation.

along with other polysaccharides (e.g., celluloses, chitosan, etc.), is indeed considered an extremely promising material in the viewpoint of circular economy and in substituting petroleum-derived polymers. [3] Among others, one of the reasons for alginate success is its unique capability to bind different cations leading to stable and tailor-made hydrogels. [4] Owing to its biocompatibility, biodegradability, and nontoxicity, in the past decades alginate has been vastly explored for drug and gene delivery, tissue engineering, and wound healing applications. [5] In this sense, the recent nanotechnological advances have allowed the fabrication and exploitation of alginate-based nanostructured materials with completely new capabilities. Thereby, it is not surprising that nowadays a broad variety of alginate-based nanomaterials with various shapes, sizes, and compositions are prepared via different approaches, including controlled gelation, electrospinning and electrospraying, self-assembly, phase-separation, and micro fluidics., All these nanostructures hold the potential to interact with living organisms much more efficiently with respect to bulk systems, hence leading to specific functionalities at the same time avoiding the occurrence of toxicity issues. [6][7][8] Despite the use of alginate-based nanomaterials has been reviewed in the past, the focus has been commonly pointed to their generic properties and applications. Conversely, this review attempts to provide a more specific and comprehensive summary limited to the most recent advances in the fabrication and use of two of the most common and promising alginatebased nanomaterials, namely nanoparticles (NPs) and electrospun nanofibers (NFs), with a focus on biomedical and pharmaceutical applications.
