Background: The orodispersible films (ODFs) are ultra-thin films that dissolve rapidly within the oral cavity. These can be used to carry vitamins, nutraceutical compounds and drugs. Generally, its formulated with synthetic polymers and artificial colorants. Due to growing consumer interest in healthier products, it is necessary to study the use of natural materials for ODF production, such as fruit juices and natural extracts. In this study, ODFs were prepared using pectin, jambolan juice and extracts of guaco, pomegranate, clove or cinnamon in different concentrations. The impact of the addition of natural extracts in different concentrations was evaluated by the following properties of the films: swelling, disintegration time, tensile strength, elongation at break, Young's modulus, work of adhesion, visual and sensory. Results:The films showed a visually homogeneous purple color, slowly disintegrating (from 1.1 AE 0.1 to 2.2 AE 0.1 min), flexible, and adherent enough independently of the extract used. The anthocyanin present in the jambolan juice (6.58 AE 0.02 mg g À1 ) contributed higher antioxidant capacity shown by the ODF (198-292 μMol TEg À1 ). The sensory evaluation reveals that the films presented adequate acceptance for a new kind of product. Conclusion:The results indicate the use of jambolan juice and natural extracts is suitable for the formulation of ODFs as a healthy product, maintaining technological properties.

Objective: Thin film (TF) is an oral mucosal drug delivery system that disintegrates and dissolves the drug rapidly when placed on any oral mucosal tissue. This work aimed to develop and characterize TF containing carbinoxamine maleate (CM) for the symptomatic treatment of seasonal and perennial allergies and other allergic conditions. Methods: Three optimized batches were developed by solvent-casting method with different ratios of blends of hydroxypropyl methylcellulose (HPMC) E50LV and polyvinylpyrrolidone (PVP) K12 or K85-95 as mucoadhesive film-forming polymers. The produced TFs were subjected to physicochemical and mechanical characterization. Results: The weight and thickness of TF were in the range of 40.47 mg ± 0.49 to 65.70 mg ± 1.51 and 50.80 μm ± 1.11 to 95.20 μm ± 2.50, respectively. The percent drug load was found to be within the acceptable limit for content uniformity. Within 15 minutes, the amount of CM released from thin films of Batch 3 was 81.04% ± 1.91 which was significantly slower than Batch 1 and Batch 2. The disintegration of TFs was achieved in less than 180 seconds. Results of mechanical testing revealed high values of Young’s modulus with no significant difference between batches. The incorporation of blends of HPMC E50 LV and PVP K85-95 at a ratio of 1:1 notably decreased tensile strength and elongation at break. Conclusion: This work demonstrates the successful development of TF as an adequate delivery system for the administration of CM. The selection of the film-forming polymer type, grade, and ratio can impact the characteristics of the TF and modify the delivery of CM. Microneedles are a transdermal drug delivery system in which the needle punctures the epithelium to deliver the drug directly to deep tissues, thus avoiding the influence of the first‐pass effect of the gastrointestinal tract and minimizing the likelihood of pain induction. Hydrogel microneedles are microneedles prepared from hydrogels that have good biocompatibility, controllable mechanical properties, and controllable drug release and can be modified to achieve environmental control of drug release in vivo. The large epithelial tissue in the oral cavity is an ideal site for drug delivery via microneedles. Hydrogel microneedles can overcome mucosal hindrances to delivering drugs to deep tissues; this prevents humidity and a highly dynamic environment in the oral cavity from influencing the efficacy of the drugs and enables them to obtain better therapeutic effects. This article analyzes the materials and advantages of common hydrogel microneedles and reviews the application of hydrogel microneedles in the oral cavity.
