Abstract. Postpartum uterine infections affect ovarian function and delay ovulation in cattle. As dietary fats can affect immune cell function, we investigated the influence of prepartum diets on postpartum uterine inflammatory status (UIS) as assessed 25 AE 1 days postpartum by endometrial cytology (normal: #8% polymorphonuclear cells (PMN) vs subclinical endometritis (SCE): .8% PMN) and associations between SCE, pro-and anti-inflammatory cytokine gene expression and ovarian function. During the last 5 weeks of gestation, dairy cows received a diet supplemented with 8% rolled sunflower (n ¼ 10) or canola seed (n ¼ 9) or no oilseed (n ¼ 9). Ovaries were scanned until 35 days postpartum. Prepartum diets did not influence SCE, but a preovulatory-size follicle developed sooner (P # 0.05), the interval to first ovulation was shorter and the proportion of cows ovulating within 35 days postpartum was greater in the sunflower seed group. Although mRNA expression of cytokines was not affected by diet, cows with SCE had higher (P # 0.05) expression of interleukin-1b (IL1B), interleukin-8 (CXCL8), IL10 and tumour necrosis factor-a (TNF) than normal cows. The interval (mean AE s.e.m.) from calving to preovulatory-size follicle was shorter (P # 0.05) in normal (13.2 AE 0.9 days) than SCE cows (18.7 AE 1.4 days). In summary, a prepartum diet supplemented with sunflower seed positively influenced postpartum ovarian function without affecting UIS or pro-and anti-inflammatory cytokine gene expression in endometrial cells.

Transmembrane mucins (TMs) are extremely large, complex glycoproteins that line the apical surfaces of simple epithelia including those of the female reproductive tract. TMs provide a physical barrier consistent with their role as part of the innate immune system. This barrier function must be overcome in the context of embryo implantation to permit blastocyst attachment. Three major TMs have been identified in uterine epithelia of multiple species: MUC1, MUC4, and MUC16. MUC1 has been found in all species studied to date, whereas expression of MUC4 and MUC16 have been less well studied and may be species specific. The strategies for removing mucins to permit embryo attachment also vary in a species-specific way and include both hormonal suppression of TM gene expression and membrane clearance via cell surface proteases. Studies emerging from the cancer literature indicate that TMs can modulate a surprisingly wide variety of signal transduction processes. Furthermore, various cell surface proteins have been identified that bind either the oligosaccharide or protein motifs of TMs suggesting that these molecules may support cell attachment in some contexts, including trophoblast interactions with cells of the immune system. The intimate association of TMs at sites of embryo-maternal interaction and the varied functions these complex molecules can play make them key players in embryo implantation and placentation processes.

Declining in reproductive efficiency is a worldwide problem, although remarkable progress has been made in reproductive techniques including artificial insemination programs, estrous cycle synchronization, and effective embryo production. One means of solving this problem is to select breeding cows based on highly precise diagnosis for high reproductive ability, including ovarian and uterine function. Many recent studies have documented the regulation of uterine function, with the results suggesting that said functions might be an effective diagnostic index for reproductive performance. Therefore, the production of oxytocin-induced prostaglandin F2α and epidermal growth factor in bovine endometrium, such as the physiological reaction in vivo, may be useful in detecting repeat breeder cows. Although the diagnosis is possible using relatively simple techniques such as biopsy, culture, and hormone assay, it is difficult to apply such diagnosis to the field conditions. However, further improvements in the exhaustive gene expression analysis of uterine function in repeat breeder or low fertility cows, so as to determine the genes associated with physiological reactions, could lead to new diagnostic indexes resulting in improved reproductive efficiency.
