MTORC1-mediated NRBF2 phosphorylation functions as a switch for the class III PtdIns3K and autophagy

Abstract: NRBF2/Atg38 has been identified as the fifth subunit of the macroautophagic/autophagic class III phosphatidylinositol 3-kinase (PtdIns3K) complex, along with ATG14/Barkor, BECN1/Vps30, PIK3R4/p150/Vps15 and PIK3C3/Vps34. However, its functional mechanism and regulation are not fully understood. Here, we report that NRBF2 is a fine tuning regulator of PtdIns3K controlled by phosphorylation. Human NRBF2 is phosphorylated by MTORC1 at S113 and S120. Upon nutrient starvation or MTORC1 inhibition, NRBF2 phosphoryla… Show more

“…The other complex I-specific subunit, Atg38, helps to maintain the integrity of complex I [33]. NRBF2 is an apparent orthologue of budding yeast Atg38 in mammals, but its exact roles remain unresolved [34][35][36][37][38][39].…”

“…Dysfunction of autophagy has been implicated in a broad spectrum of human diseases including cancers, neurodegeneration, infectious diseases, metabolic diseases and aging [39]. Autophagy can be activated under stress conditions such as starvation, endoplasmic reticulum (ER) stress, and organelles damage and pathogen infection.…”

“…NRBF2 is a regulator of autophagy, a cellular process for disassembly of dysfunctional cytosolic components . In its phosphorylated form, NRBF2 inhibits autophagy, while promoting it in its unphosphorylated form . Intact autophagy, in turn, was recently reported to confer protection against autoimmune disorders such as lupus nephritis .…”
