MtlR negatively regulates mannitol utilization by Vibrio cholerae

Abstract: The phosphoenopyruvate:carbohydrate phosphotransferase system (PTS) enables Vibrio cholerae – and other bacteria – to recognize and transport exogenous carbon sources for energy, including the six-carbon sugar alcohol, mannitol. The mannitol-specific PTS transporter is encoded by mtlA and its expression is expected to be regulated by the putative repressor encoded by the mtlR gene. Here, we show that mtlR overexpression inhibits V. cholerae growth in medium supplied with mannitol as the sole carbon source and … Show more

“…Mannitol is a polyol produced by marine algae and is the most abundant hexitol found in fungi, algae, and plants [ 25 , 26 ]. Mannitol metabolism has been intensively studied in marine bacteria, such as Vibrio cholerae and Zobellia galactanivorans [ 26 , 27 ]. Furthermore, mannitol is widely used in food, pharmaceutical, medical and chemical industries [ 28 ].…”

“…The MtlR repressor protein, which is encoded in the mtlADR operon itself, and MtlS appear to operate independently from each other (59,61). MtlA was shown to activate biofilm formation suggesting that mannitol serves as extracellular signal for V. cholerae to colonize beneficial habitats (62).…”

“…cholerae which encodes three genes related to the transport and metabolism of mannitol: mtlA (encoding the mannitol-specific enzyme IIABC component of the phosphotransferase system [PTS]), mtlD (a mannitol-1-phosphate dehydrogenase), and mtlR (a transcriptional repressor of mtlA ) (Fig. 1A) (2224). Mannitol is one of the most abundant and widely distributed natural sugar alcohols and the primary photosynthetic product of brown algae (25, 26).…”

“…The global regulator cAMP receptor protein (CRP) is a transcriptional activator of mtlA (29). Opposing the activity of CRP, MtlR acts as a transcriptional repressor of mtlA (23). Studies concerning the regulation of the mtl locus provide a model for maximal mtlA transcription that relies on two conditions: high cAMP-CRP activity and low MtlR activity (23, 29).…”
