MRI of perfluorocarbon emulsion kinetics in rodent mammary tumours

Abstract: Perfluorocarbon (PFC) emulsions can be imaged directly by fluorine-19 MRI. We developed an optimized protocol for preparing PFC droplets of uniform size, evaluated use of the resulting droplets as blood pool contrast agents, studied their uptake by tumours and determined the spatial resolution with which they can be imaged at 4.7 T. Perfluorocarbon droplets of three different average sizes (324, 293 and 225 nm) were prepared using a microemulsifier. Images of PFC droplets with good signal-to-noise ratio were a… Show more

“…PFCE 2g, Lipoïd 0.12g, glycerin 0.09g and deionized water 2.91g, larger particles were obtained with a hydrodynamic diameter (Z-ave) ~ 250nm and a higher polydispersity index (PdI ~ 0.25). Interestingly, those nanoparticles are however comparable in size with those obtained using the high-energy process described by Fan et al [35].…”

“…In the literature, sonication in combination with high pressure is often used to obtain PFCELNCs [35,36]. In our case, sonication at atmospheric pressure was used to improve the dispersion of the oil and water phases before the phase-inversion process.…”

“…Since then, 19 F imaging has also been utilized for cell-tracking [39,40], tumor imaging [41], and especially in imaging of inflammation [42][43][44][45][46][47]. The C-F bond in perfluorocarbons is amongst the strongest chemical bonds known to man; consequently, PFCs are some of the most stable and inert molecules available for any application.…”

“…Despite several examples of successful 19 F in vivo imaging [39][40][41][42][43][44][45][46][47], the MR experiment is inherently limited by low signal sensitivity, and fluorine MRI is often hindered by low SNR due to the limited number of fluorine nuclei that can be delivered to an area of interest. In order to obtain reasonable voxel sizes during an in vivo imaging session, dozens or hundreds of averaging scans are often used, resulting in lengthy examination times.…”
