MR extracellular volume mapping and non-contrast T1ρ mapping allow early detection of myocardial fibrosis in diabetic monkeys

Abstract: Objective To detect diffuse myocardial fibrosis in different severity levels of left ventricular diastolic dysfunction (DD) in spontaneous type 2 diabetes mellitus (T2DM) rhesus monkeys. Methods Eighteen spontaneous T2DM and nine healthy monkeys were studied. Echocardiography was performed for diastolic function classification. Cardiac magnetic resonance (CMR) imaging was performed to obtain extracellular volume fraction (ECV) maps and T1ρ maps at two different spin-loc… Show more

“…after myocardial infarction (MI), can cause harmful remodeling of the myocardium, ultimately leading to heart failure [ 3 6 ]. At cellular level, cell death is followed by an increase in extracellular space, which affects the interaction between free water and macromolecules [ 7 , 8 ]. Applying advanced imaging tools in both animal models as well as patient care has led to greatly increased knowledge [ 9 11 ], proving that even subtle scar formation has important diagnostic, prognostic and therapeutic clinical implications [ 12 ].…”

“…Thus, T 1ρ provides a more universal indicator and is only restricted by specific absorption rate (SAR) limitations. Recently, a study in diabetic monkeys was able to demonstrate T 1ρ dispersion within SAR limitations as a myocardial index for the early detection of diffuse fibrosis [ 8 ]. This study clearly demonstrates that the research of T 1ρ in animal models can provide important insights for specific development of clinical CMR sequences.…”

“…Application of T1ρ mapping to CMR has been reported in multiple studies [ 5 8 ], mostly to discriminate between infarct and healthy myocardium in swine, mouse and monkey models. These studies raise the interesting question of whether endogenous T1ρ mapping could be a useful adjunct to late gadolinium enhancement (LGE) imaging.…”

“…The results showed similar associations between DCM and myocardial dysfunction as STE, but with higher costs. These studies go beyond the purpose of our review 7,37,38,43,53‐63 . The principal advantages and limitations of CMR compared to STE and TDI are showed in Table 2.…”
