Movement of yeast 1,3‐β‐glucan synthase is essential for uniform cell wall synthesis

Abstract: Background:  The cell wall has an important role in maintaining cell shape. In the budding yeast Saccharomyces cerevisiae, the major filamentous component of the cell wall responsible for its rigidity is 1,3‐β‐glucan and is synthesized by 1,3‐β‐glucan synthase (GS), localized on the plasma membrane. Results:  Observations of green fluorescent protein (GFP)‐conjugated Fks1p, a catalytic subunit of GS, revealed that it is co‐localized with cortical actin patches and moves on the cell surface at the sites of cell… Show more

“…Other factors affect ␤-1,3-glucan synthesis by regulation of the catalytic subunit of GS. Movement of Fks1p driven by actin is required for the construction of a uniform and solid cell wall (Utsugi et al, 2002). Transcription of FKS2 is up-regulated in response to cell wall stress induced by heat, cell wall mutations, and cell wall-perturbing agents (Zhao et al, 1998;de Nobel et al, 2000;Lagorce et al, 2003;Garcia et al, 2004).…”

“…For example, mutations in both FKS1 and FKS2 result in lowered b1,6-glucan synthesis (Dijkgraaf et al 2002). Fks1 is localized to the plasma membrane at sites of polarized growth and cell wall remodeling throughout the cell cycle, and this localization coincides with that of actin patches (Qadota et al 1996;Dijkgraaf et al 2002;Utsugi et al 2002). Fks1 transits the secretory pathway because it accumulates intracellularly in vesicular transport mutants and its activity is sensitive to phytosphingosine levels in the ER (El-Sherbeini and Clemas 1995; Abe et al 2001;File S7).…”
