Mouse papillomavirus type 1 (MmuPV1) DNA is frequently integrated in benign tumors by microhomology-mediated end-joining

Abstract: MmuPV1 is a useful model for studying papillomavirus-induced tumorigenesis. We used RNA-seq to look for chimeric RNAs that map to both MmuPV1 and host genomes. In tumor tissues, a higher proportion of total viral reads were virus-host chimeric junction reads (CJRs) (1.9‰ - 7‰) than in tumor-free tissues (0.6‰- 1.3‰): most CJRs mapped to the viral E2/E4 region. Although most of the MmuPV1 integration sites were mapped to intergenic regions and introns throughout the mouse genome, integrations were seen more tha… Show more

“…Low-risk HPV E5, E6, and E7 proteins have weaker or altered functions compared to their high-risk counterparts [ 115 ]. As in high-risk HPVs, MmuPV1 can integrate into the host genome in diseased tissues [ 116 ]. Low-risk HPV genomes generally do not integrate in RRP [ 91 ].…”

“…In support of the “hit and runs” mutagenesis hypothesis, deletion of E6/E7 after these lesions form did not affect cancer growth [ 29 ]. Rodent papillomaviruses resembling beta HPV were used to aid the study of cutaneous HPV biology in vivo [ 62 , 63 , 64 , 65 ]. Mastomys natalensis papilloma virus (MnPV) promotes cSCC in immunodeficient rats following UVB exposure [ 57 ].…”

“…Integration probably occurs frequently, particularly in a genetically unstable environment, but only those integration events that provide cells with a selective growth advantage drive clonal expansion and oncogenesis [ 102 , 103 ]. In fact, MmuPV1 (Mus musculus papillomavirus 1) genomes are frequently integrated in benign mouse tumors [ 104 ].…”
