Mouse MutS-like protein Msh5 is required for proper chromosome synapsis in male and female meiosis

Abstract: Members of the mammalian mismatch repair protein family of MutS and MutL homologs have been implicated in postreplicative mismatch correction and chromosome interactions during meiotic recombination. Here we demonstrate that mice carrying a disruption in MutS homolog Msh5 show a meiotic defect, leading to male and female sterility. Histological and cytological examination of prophase I stages in both sexes revealed an extended zygotene stage, characterized by impaired and aberrant chromosome synapsis, that was… Show more

“…Nevertheless, C. elegans msh-5 mutants are able to complete meiosis and gametogenesis with high efficiency and produce embryos without undergoing germ cell apoptosis like their murine counterparts. 59,60 It is likely that msh-5 mutant cells escape apoptosis because they are still proficient in repairing dsDNA breaks, but in a way that does not lead to crossovers between homologous chromosomes. 58 …”

“…In mice MSH4 and MSH5 have an early role in DSB repair. Indeed meiocytes arrest at zygonema/early pachynema [42][43][44][45] precluding a cytological analysis of CO formation. However, the co-localization (on meiotic chromosome spreads) of MSH4 with a marker of pro-CO and CO events (MLH3 and MLH1 respectively) [46,47] and the ability of hMSH4/hMSH5 heterodimer to bind Holliday Junctions in vitro [48], indicates that these proteins have also a role in late recombination.…”
