Mouse gastric mucin: cloning and chromosomal localization

Abstract: Mucins protect gastric epithelium by maintaining a favourable pH gradient and preventing autodigestion. The purpose of this study was to clone a mouse gastric mucin which would provide a foundation for analysis of mucin gene regulation. Mucin was purified from the glandular portion of gastric specimens and deglycosylated by HF solvolysis. Antibodies against native and deglycosylated mouse gastric mucin (MGM) were raised in chickens. Screening of a mouse stomach cDNA library with the anti-(deglycosylated MGM) a… Show more

“…Muc2, a large secreted mucin, is a goblet cell product that is found mainly in the small intestine and colon (17). Muc3 is another intestinal mucin that has been cloned partially by two different groups of investigators (18,19). Probes for M2 and for rMuc176 both hybridize to a 9-kb RNA from small intestine and colon.…”

“…In the small intestine and colon, Muc1 is produced at very low levels by enterocytes. Muc2 and Muc5ac are secreted glycoproteins, whereas Muc3 has been described recently as a transmembrane mucin (16)(17)(18)(19). In addition to numerous tandem repeats, these mucins have cysteinerich domains that are responsible for self-dimerization.…”

“…Gastric mucin and cathepsin E are produced by pit cells (Shekels et al, 1995;Fukamachi et al, 1994), and gastric H + ,K + -ATPase and pepsinogen are expressed in parietal (Shull and Lingrel, 1986;Smolka et al, 1983) and zymogenic cells (Fukamachi et al, 1994), respectively. The cells were tested for the mucosal cell-specific mRNA expression using a RT-PCR.…”
