Mouse adenovirus type 1 infection of natural killer cell-deficient mice

Abstract: Natural killer (NK) cells contribute to the initial nonspecific response to viral infection, and viruses exhibit a range of sensitivities to NK cells in vivo. We investigated the role of NK cells in infection of mice by mouse adenovirus type 1 (MAV-1) using antibody-mediated depletion and knockout mice. MAV-1 causes encephalomyelitis and replicates to highest levels in brains. NK cell-depleted mice infected with MAV-1 showed brain viral loads 8-20 days p.i. that were similar to wild-type control non-depleted m… Show more

“…This has so far been investigated both in vitro and in vivo using the wildtype mouse adenovirus type‐1 (MAV‐1) which causes hemorrhagic encephalitis in mice. To recover from acute MAV‐1 infection, mice need a robust antibody response . The ability of anti‐MAV‐1 serum to neutralize MAV‐1 was assessed in cells derived from wildtype or TRIM21 knockout mouse embryonic fibroblasts, and a significant proportion of neutralization was TRIM21‐dependent .…”

“…Interestingly, depletion of the NK response in mice does not affect their sensitivity to murine Ad type (Welton et al, 2008). With human Ad5, either expression of E1A or breakthrough expression from a E1 vector can stimulate cell surface expression of ligands for the powerful NK cell activity receptor NKG2D; however, this is countered by E3 19K (Routes et al, 2005;Tomasec et al, 2007;McSharry et al, 2008).…”

“…Passive transfer of neutralizing antibodies is sufficient to protect btk −/− mice and reduce viremia in a RAG −/− strain (7). In contrast, depletion of natural killer cells has a limited affect on both viremia and survival (8), whereas mouse strains lacking natural killer cells, CD4…”
