Motor neuron boutons remodel through membrane blebbing

Abstract: Wired neurons form new presynaptic boutons in response to increased synaptic activity, but the mechanism by which this occurs remains uncertain. The neuromuscular junction (NMJ) is a synapse formed between motor neurons (MNs) and skeletal muscle fibers and is critical for control of muscle contraction. Because Drosophila MNs have clearly discernible boutons that display robust structural plasticity, it is the ideal system in which to study bouton genesis. Here we show using ex-vivo and by live imaging that in … Show more

“…this paradigm, spaced high potassium stimulation promotes acute formation of nascent ghost boutons (Ataman et al, 2008;Korkut et al, 2013;Piccioli and Littleton, 2014). These are likely transient structures, and thus are not directly comparable to developmentally arrested ghost boutons such as those that are found in evi mutants (Fernandes et al, 2021). However, to avoid the confounding presence of these immature boutons, we explored the activity-dependent synaptic growth paradigm on muscle 4, where the Tsg101 KD animals do not have significantly more ghost boutons than controls under baseline conditions.…”
