Mosquito Vitellogenin Genes: Comparative Sequence Analysis, Gene Duplication, and the Role of Rare Synonymous Codon Usage in Regulating Expression

Abstract: Comparative sequence analysis of mosquito vitellogenin (Vg) genes was carried out to gain a better understanding of their evolution. The genomic clones of vitellogenin genes were isolated and sequenced from all three subfamilies of the family Culicidae including Culicinae (Aedes aegypti, Ochlerotatus atropalpus, Ae. polynesiensis, Ae. albopictus, Ochlerotatus triseriatus and Culex quinquefasciatus), Toxorhynchitinae (Toxorhynchites amboinensis), and Anophelinae (Anopheles albimanus). Genomic clones of vitellog… Show more

“…Recently, in silico whole‐genome analysis isolated several new Vg gene sequences from several mosquito species including Anopheles albimanus (Wiedemann), Ae. aegypti , Aedes polynesiensis (Marks), Aedes albopictus (Skuse), Ochlerotatus atropalpus (Coquillett), Ochlerotatus triseriatus (Say), Culex pipiens (L.) and Toxorhynchites amboinensis (Doleschall) (Isoe & Hagedorn ). Nevertheless, it remains unclear why some insects harbor multiple Vg genes.…”

“…However, the level of Vg RNA was too low to support the process of egg development. This may be because of the presence of certain rare synonymous codons that might have initiated the process of vitellogenesis after accumulating at the 5' end in the absence of any external amino acid supply 23 . Also, as described earlier, the induction of Vg expression after a blood meal requires the Target of Rapamycin (TOR) pathway and ecdysone signaling 24,25 .…”

“…Mosquitoes contain ~100 ovarioles per ovary, which are composed of primary and secondary follicles along with a germarium. Mosquito ovarian follicles develop synchronously throughout oogenesis (Hagedorn et al, 1977;Lea et al, 1978;Clements and Boocock, 1984;Raikhel et al, 2002;Uchida et al, 2004;Isoe and Hagedorn, 2007;Clifton and Noriega, 2011). The spherical follicle gradually transforms into an ellipsoid form by accumulating vitellogenin yolk proteins.…”
