Mosquito species (Diptera, Culicidae) in three ecosystems from the Colombian Andes: identification through DNA barcoding and adult morphology

Abstract: Colombia, one of the world’s megadiverse countries, has a highly diverse mosquito fauna and a high prevalence of mosquito-borne diseases. In order to provide relevant information about the diversity and taxonomy of mosquito species in Colombia and to test the usefulness of DNA barcodes, mosquito species collected at different elevations in the departments of Antioquia and Caldas were identified combining adult morphology and barcode sequences. A total of 22 mosquito species from eight genera were identified us… Show more

“…NJ trees using K2P substitution model are the most widely constructed phylogenetic trees used in many barcoding studies, especially on mosquitoes, to obtain more accurate recognition of the targeted group [3, 6, 9, 49]. Accordingly, the same methodology was applied during this study to increase the reliability of the results.…”

“…MEGA v 6.06 program (Tamura, Stecher, Peterson, Filipski, & Kumar, ) was used for sequences alignment, for searching for stop codons and for the calculation of intra‐ and interspecific genetic distances using the Kimura two parameters distance model (K2P) (Kimura, ; Tamura et al, ). To provide a graphic representation of the grouping pattern between the different species a K2P distance similarity tree was constructed using the Neighbor‐Joining model (NJ) with Bootstrap support values calculated with 1,000 replicates (Laurito et al, ; Rozo‐López & Mengual, ; Saitou & Nei, ).…”

“…Recent studies indicate that it is necessary to keep identifying mosquito species in different regions of the world through the use of traditional taxonomy and DNA sequences, to achieve a better estimation of mosquito biodiversity (Ashfaq et al, ; Azari‐Hamidian et al, ; Batovska, Blacket, Brown, & Lynch, ; Cywinska, Hunter, & Hebert, ; Rozo‐López & Mengual, ; Versteirt et al, ; Wang et al, ; Weeraratne, Surendran, & Parakrama Karunaratne, ). The combined use of DNA and morphology allowed to analyze a greater number of species worldwide and to discover cryptic speciation in taxa of medical importance such as culicides (Ashfaq et al, ; Hebert & Gregory, ; Laurito, Oliveira, Almirón, & Sallum, ; Torres‐Gutiérrez et al, ; Wang et al, ).…”

“…Standardized mitochondrial fragments from COI (DNA barcodes) have been used to identify mosquitoes (Cywinska et al 2006; Harrison et al 2012; Kumar et al 2007; Ruiz-Lopez et al 2012), including species from the Colombian Andes (Rozo-Lopez & Mengual 2015). Also, in Colombia, DNA barcodes were used to distinguish An.…”
