Mosaic Genetic Screen for Suppressors of the de2f1 Mutant Phenotype in Drosophila

Abstract: The growth suppressive function of the retinoblastoma (pRB) tumor suppressor family is largely attributed to its ability to negatively regulate the family of E2F transcriptional factors and, as a result, to repress E2F-dependent transcription. Deregulation of the pRB pathway is thought to be an obligatory event in most types of cancers. The large number of mammalian E2F proteins is one of the major obstacles that complicate their genetic analysis. In Drosophila, the E2F family consists of only two members. The… Show more

“…Moreover, it is surprising that mutations in Caf1 were not identified in previous Drosophila modifier screens involving PcG or Rb pathway members (Ambrus et al, 2009;Janody et al, 2004;Steele et al, 2009). We propose that the link between sens and Caf1 is due to the role of Caf1 in PcG-mediated silencing.…”

“…Interestingly, deprivation of SLIMP has been recently identified in a genetic mosaic screen as a compensator of the cell growth arrest phenotype caused in Drosophila cells by the inactivation of transcription factor dE2F1 (46). Although the significance of this observation is still unclear, it suggests to us that the mitochondrial role of SLIMP, or a product of its func- tion, may be acting as a signal between mitochondria and nuclear transcription factors that regulate cell proliferation.…”

“…dsRNA knockdown was done as described in Ambrus et al (2009). Three days after initial soaking, cells were split and the following day, cells were transfected with the X-tremeGENE HP transfection reagent (Roche) according to manufacturer's protocol.…”
