ABSTRAKIkan belut sawah (Monopterus albus) merupakan spesies ikan air tawar ekonomis penting yang potensial untuk dikembangkan secara berkelanjutan. Spesies ini dianggap sebagai spesies bermasalah karena karakternya yang komplek serta morfologinya yang mirip dengan Monopterus cuchia dan Monopterus javanensis yang sering kali menghantarkan dilema taksonomi. Kepastian taksonomi merupakan informasi dasar penting dalam pengelolaan perikanan secara berkelanjutan, studi keragaman morfometrik, dan genetik merupakan metode umum yang digunakan untuk identifikasi spesies maupun stok. Penelitian ini untuk memvalidasi, identifikasi, serta evaluasi hubungan kekerabatan spesies belut sawah asal empat populasi (Indramayu, Subang, Tasikmalaya, dan Garut) di Jawa Barat. Analisis keragaman morfometrik dilakukan berdasarkan pengukuran 19 karakter morfometrik, sedangkan analisis keragaman genetik dilakukan menggunakan metode PCR-Sekuensing mtDNA gen COI. Hasil analisis diskriminan stepwise menghasilkan 7 rasio karakter morfometrik yang berpengaruh terhadap pengelompokan populasi dan dua fungsi diskriminan pertama menyumbang 84,2% dari total variasi. Keragaman genetik ditunjukkan melalui penjajaran 14 sekuen gen COI yang menghasilkan 11 haplotipe spesies ingrup dari 109 situs nukleotida variasi. Hasil analisis klaster dan pohon filogenetik menunjukkan bahwa ikan belut sawah dari empat populasi penelitian mengelompok ke dalam dua klaster utama. Klaster pertama terdiri atas populasi Indramayu dan Subang, klaster kedua terdiri atas populasi Tasikmalaya dan Garut. Pengelompokan tersebut membuktikan bahwa populasi belut sawah di Jawa Barat secara morfometrik dan genetik bervariasi di antara populasi dan menghantarkan pada dugaan bahwa sedikitnya ada dua spesies kriptik dari M. albus di Jawa Barat.Kata kunci: Jawa Barat, keragaman genetik, keragaman morfometrik, Monopterus albus ABSTRACT Swamp eel (Monopterus albus) is an economically important freshwater fish which has the potential to sustainable development. This species is considered as one of the taxonomically problematic spesies due to its complex characters and similar morphology to Monopterus cuchia and Monopterus javanensis that often lead to taxonomic dilemmas. Taxonomic certainty is an essential basis information in sustainable fisheries management, studies on morphometric characters, and genetic variability is a method which commonly used for species or stock identification. This study was aimed to validate the taxonomy certainty and evaluate the kinship of swamp eel from four populations (Indramayu, Subang, Tasikmalaya, and Garut distrik) in West Java. Morphometric analysis was conducted by measurement of 19 characters, whereas genetic analysis was conducted using PCR-Sequencing method of COI gene.
