Morphology and phylogeny of Pseudorobillarda eucalypti sp. nov., from Thailand

Abstract: Pseudorobillarda is a coelomycete genus of Dothideomycetes with appendaged conidia and 15 species epithets. In this study, we isolated four strains of Pseudorobillarda from dead leaves in Thailand. DNA sequence data generated from the large subunit (28S) ribosomal DNA (nuLSU) gene was used in phylogenetic studies. The phylogenetic trees generated indicate that Pseudorobillarda form a distinct lineage in Dothideomycetes that may eventually require separate family status. The Pseudorobillarda strains comprised t… Show more

“…The specimens were divided into two parts. The first part was used for morphological study and single spore isolations prepared following the methods described in Chomnunti et al [ 11 ] and Tangthirasunun et al [ 12 13 ]. The colonies were transferred to water agar and incubated at room temperature to promote sporulation.…”

“…Primers ITS1 and ITS4 [ 17 ] and LROR and LR7 [ 18 ] were used to amplify the ITS and part of the LSU rRNA genes. PCR reaction mixtures and amplification conditions were as described by Tangthirasunun et al [ 12 13 ]. PCR products were checked on 1% agarose electrophoresis gels stained with ethidium bromide [ 19 ] and sequenced by Beckman Coulter Genomics (Danvers, MA, USA; Grenoble, France).…”

“…Notes: Pseudorobillarda is a coelomycetous genus, characterized by hyaline conidia with divergent appendages (Nag Raj 1993). The placement of Pseudorobillarda is unstable as it either clustered with Pleosporales (Rungjindamai et al 2012), or nested between Tubeufiales and Mytilinidiales (Tangthirasunun et al 2014). Recently, it was placed in a newly established family Pseudorobillardaceae which resides in Minutisphaerales (Crous et al 2018a(Crous et al , 2019b.…”

“…Notes: The members of this genus can be saprobic, pathogenic and endophytic and are known from temperate and tropical countries such as Argentina, Canada, Cuba, Germany, India, Nigeria, South Africa, Thailand, UK, Ukraine, and the USA (Petrini 1986;Nag-Raj 1993;Bianchinotti 1997;Vujanovic and St-Arnaud 2003;Plaingam et al 2005;Tangthirasunun et al 2014;Crous et al 2018a). This genus occurs on both living and dead leaves, stems or bark and is also commonly found in soil (Petrini 1986;Vujanovic and St-Arnaud 2003;Kadowaki et al 2014).…”

“…The revised descriptions and illustrations of Pseudorobillarda, based on morphological characteristics are available in Plaingam et al (2005). No sexual morph is linked to Pseudorobillarda (Hyde et al 2011;Wijayawardene et al 2012Wijayawardene et al , 2017aTangthirasunun et al 2014).…”
