A comprehensive account on standardization of herbal drug Nigella sativa L. (Tukhm-e-Kalonji) by using microscopic as well as pharmacognostic parameters. In the field of herbal medicines, the main issues are quality, purity, and effectiveness, as in many cases herbal drugs are knowingly or unknowingly substituted or adulterated with similar species or varieties. In herbal market the seeds of N. sativa are commonly adultered with seeds of Allium cepa L. intentionally or accidentally due to their similar morphology. In the present study, the microscopic characterization of herbal drug was done by morphological, palynological, and anatomical features. A great diversity was found in anatomical features of two species, as irregular epidermal cells, actinocytic stomata, and nonglandular trichomes were found in N. sativa while in A. cepa epidermal cells were rectangular in shape, stomata type was paracytic, and trichomes were absent. Pharmacognostic characterization was made by fluorescence analysis and physiochemical parameters. Physicochemical parameters like moisture content, total ash, acid insoluble ash, water soluble ash, and water insoluble ash were also evaluated. The above parameters, being reported to the first time for the studied plant species, and are significant towards establishing the microscopic and pharmacognostic standards for future identification and authentication of genuine herbal drug. (Tukhm-e-Kalonji).• Pharmacognostic techniques are potentially significant for the standardization of herbal drug Nigella sativa in comparison with its adulterant.• Useful for preparation of standards for herbal pharmacopeia.

Scanning microscopic imaging has become a valuable research tool in micromorphology with improved techniques playing an important role in analysing the ultrastructure of leaf specimens. The foliar epidermal anatomy of 25 selected poisonous plants with special emphasis on stomata and trichomes was reported using microscopic techniques, for instance, light micrographs (LMs) and scanning micrographs (SEMs). This study aimed to investigate micromorphologies of studied species that are helpful for the identification of poisonous plants. Plants were collected, pressed, dried, identified and then analysed for microscopic study. For making microscopic slides, 1 or 2 leaves were taken in a test tube and dipped in 30% nitric acid and 70% lactic acid for few minutes, and then placed on petri plates for separating the epidermis. Numerous quantitative and qualitative foliar anatomical features of adaxial and abaxial surfaces, including epidermal cell shapes, stomata size, subsidiary cell size, the pattern of the anticlinal wall, the morphology of the stomatal complex and trichome diversity, were examined. A small number of the considered species had anomocytic and anisocytic stomata; a few species had paracytic stomata, for instance, Ricinus communis, Euphorbia royleana, Buxus pilosula and Sorghum halepense; and only Ipomoea carnea had cyclocytic stomata in the studied taxa. The epidermal cells of the analysed species were irregular, while some exhibited polygonal, wavy, tetragonal and elongated cell morphologies. Overall, this study emphasises the significance of foliar micromorphology analysis as a valuable resource for identifying potentially poisonous plants and demonstrates its contribution to maintaining public welfare, thereby benefitting public health and safety.

Genus Salix is among family Salicaceae, distributing in the northern hemisphere. It is represented in Egypt by two species (Salix mucronata and Salix tetrasperma). The classification of Salix at the generic and infra-generic levels is still outstanding. We have agreed to list the Egyptian species of this genus. We collected them during field trips to most Egyptian habitats; fresh and herbarium specimens were subjected to taxonomic revision based on morphological characters; scanning electron microscope (SEM) for pollen grains; isozyme analysis using esterase and peroxidase enzymes and genetic diversity using random amplified polymorphic DNA (RAPD). We recorded that both sexes of S. mucronata existed but there were only male trees from S. tetrasperma. SEM of pollen grain revealed that there were many differences between them. Also, the application of isozyme analysis giving 12 sex indicating bands. In addition, ten RAPD primers retrieved a total of 227 amplified bands (77 for female S. mucronata, 74 for male S. mucronata and 76 for male S. tetrasperma).
