Kelinci bunting semu diperlukan untuk kegiatan yang berkaitan dengan mekanisme endokrinologi, terapi, dan transplantasi. Penelitian ini bertujuan mengetahui keberhasilan induksi bunting semu dengan metode kopulasi tiruan pada kelinci lokal. Penelitian ini mwnggunakan sembilan ekor kelinci betina lokal dan satu ekor kelinci jantan lokal, berumur 1–1,5 tahun, dan bobot badan 1,8–2,2 kg yang dibagi dalam tiga kelompok perlakuan (n= 3), yakni, kelompok kelinci yang diinjeksi dengan 0,1 mL NaCl fisiologis dan tanpa perkawinan (K1, kontrol negatif), kelinci yang mendapat induksi dengan kopulasi tiruan (K2), dan kelinci yang mendapatkan injeksi 100 IU PMSG secara intramuskuluar dan dikawinkan dengan pejantan tiga hari kemudian dan diikuti dengan injeksi 75 IU hCG secara intravena (K3, kontrol positif). Hasil penelitian diamati jumlah dan ukuran folikel serta korpus luteum dengan pemeriksaan histologis menggunakan pewarnaan HE. Jumlah folikel pada kelompok K1; K2; dan K3 masing-masing adalah 5,9 ± 1,45; 0,63 ± 0,35; dan 2,06 ± 1,50 (P <0,05). Pada kelompok K1, tidak terdapat kelinci yang ovulasi. Namun, pada kelompok K2 dan K3 seluruh kelinci berhasil ovulasi. Pada kelompok K2 dan K3 yang berhasil ovulasi, terdapat perbedaan yang signifikan (P <0,05), yaitu jumlah korpus luteum pada K2 (4,83 ± 2,65) dan pada K3 (7,63 ± 0,57), sedangkan ukuran korpus luteum pada K2 adalah 0,68 ± 0,20 dan pada K3 adalah 1,38 ± 0,16 mm. Disimpulkan bahwa kopulasi tiruan dapat menginduksi bunting semu pada kelinci lokal.AbstractPseudo-pregnant rabbits are required for activities related to endocrinology, therapy, and transplantation mechanisms. This study aims to determine the success of pseudopregnancy induction with artificial copulation methods in local rabbits. In this study, 9 local female rabbits and 1 local male rabbit aged 1–1.5 years-old and 1.8–2.2 kg body weight were used, which were divided into three treatment groups (n= 3). The rabbit in group K1 (negative control) were injected with 0.1 mL of physiological NaCl and were not mated. The rabbits in K2 were induced with artificial copulation by inserting a cotton bud of 1 cm into vagina at five o'clock in the morning, while the rabbits in K3 (positive control) received injection of 100 IU PMSG intramuscularly and mated with males three days post injection and then followed by injection of 75 IU hCG intravenously. The number and size of follicles and corpus luteum were determined by histological examination with HE staining. The number of follicles in the K1 group; K2; and K3 were 5.9 ± 1.45; 0.63 ± 0.35; and 2.06 ± 1.50, respectively (P <0.05). There was no ovulation observed at rabbit in K1 but all rabbits ovulated successfully in K2 and K3. The number of CL (4.83 ± 2.65 and 7.63 ± 0.57) and the size of CL (0.68 ± 0.20 and 1.38 ± 0.16 mm) were significantly difference (P <0.05) in groups K2 and K3, respectively. It was concluded that artificial copulation could induce pseudopregnancy in local rabbits.

Understanding the does reproductive hemodynamic changes during the estrous cycle is crucial for improving reproductive competence and fertility potential in this species. The objective of this study is to investigate the hemodynamic variations in ovarian (OA) and uterine (UA) arteries, histological and morphometric changes in ovarian and uterine tissues throughout the follicular (FP) and luteal (LP) phases in rabbits and determine estrogen (ER), progesterone (PR) receptors, and vascular endothelial growth factor (VEGF) distributions using immunohistochemistry.Fourteen adults pluriparous New Zealand rabbits were divided into rabbits at the FP (Day − 1; n = 7) and those at the LP (Day 9; n = 7). Animals were subjected to Doppler, hormonal (estrogen [E2], progesterone [P4], insulin-like growth factor [ILGF], and VEGF), histological, and immunohistochemical analyses. In LP, OA Doppler indices were significantly increased, whereas peak systolic velocity (PSV) was decreased compared with that in FP. UA Doppler indices were significantly decreased in the LP, whereas PSV was increased (P < 0.05). E2 levels were increased in the FP, whereas P4 levels were increased in the LP. The morphometric analysis of uterine tissues during the LP revealed an increase in the mean uterine endometrium length, endometrial connective tissue area percentage (%), endometrial glands number, myometrial area (%) and thickness. Furthermore, ovarian follicles and corpus luteum (CL) displayed strong positive immunoreactivity for ER, PR, and VEGF-A during both phases. The ovarian sections displayed a substantial (P < 0.05) increase in the area % of VEGF-A in the ovarian follicles during FP while in the CL during LP. Conversely, area percentage of VEGF-A immunoreactivity in the uterine luminal and glandular epithelia during the FP and LP revealed no differences. However, the number of VEGF-A–stained blood capillaries revealed an increase during LP than FP. In conclusion, this study demonstrated for the first time the changes in both ovarian and uterine arteries during two different phases of the rabbit cycle in relation to the histo-morphometric analysis and distribution of ER, PR, and VEGF-A, which regulate uterine functions that play a role in reproduction.

Comparison of four methods of inducing pseudopregnancy in rabbits

This study was conducted to compare several methods for inducing pseudopregnancy in local rabbits. The study used 25 clinically healthy rabbits of local breeds, aged 1-1.5 years, with a bodyweight of 1.8-2.2 kg. The rabbits were divided into five treatment groups (n = 5): R1, R2, R3, R4, and R5. Each group was given a different treatment. Group R1 was injected intramuscularly with 100 IU PMSG and mated with males three days later, which was followed by an intravenous injection of 75 IU hCG. Pseudopregnancy in group R2 was induced through artificial copulation by inserting a 1 cm long cotton bud into vaginal lumen at 5 a.m. Pseudopregnancy in group R3 was induced using 50 IU hCG intramuscularly. Group R4 was injected intravenously with 5 g GnRH and then intramuscularly with 100 IU PMSG and three days later with 75 IU hCG. Blood samples were collected on days 2, 4, 6, and 8 for estrogen and progesterone measurement, whereas an ovarian histological observation of all groups was performed on day 8. The results show that the highest concentration of estrogen was obtained in group R2, which was significantly different from the other groups (P <0.05), but the difference in examination time was not statistically significant (P > 0.05). Furthermore, the concentration of progesterone in all groups fluctuated at various times of the day. Progesterone concentrations were higher in group R5, in which pseudopregnancy was induced with PMSG and hCG. Group R5 showed a significant difference (P <0.05) with R2, R3, and R4, but the difference with the positive control group (R1) was not significant (P > 0.05). The numbers of corpora lutea in groups R1, R2, R3, R4, and R5 were, respectively, 6.87 ± 1.58, 6.22 ± 2.71, 6.74 ± 1.94, 5.98 ± 2.04, and 8.8 ± 1.90, and the largest diameter was obtained in R5 (1.65 ± 0.37 mm). In conclusion, the best method of inducing pseudopregnancy in local rabbits is the administration of PMSG and hCG.
