Concurrent administration of chloramphenicol (CAP) with multivitamin-haematinics complex (MHC) is a common practice to cushioning anticipated anaemic effect of CAP in most developing countries. This study investigated the mechanism involved in CAP-induced reproductive toxicity as well as the effects of its co-administration with MHC in male rats. CAP and MHC were administered orally at therapeutic doses of 28 mg ⁄ kg body-weight and 0.08 ml ⁄ kg body-weight, respectively, every 6 hr for 10 days. After exposure, while there was body-weight loss in CAP, MHC and CAP plus MHC-treated animals, there were no treatment-related changes in the absolute and relative weights of the testes in all treated groups. Alone, MHC treatment markedly decreased catalase (CAT), glutathione S-transferase (GST), and 5¢ nucleotidase (5¢ NTD) activities whereas it resulted in significant increase in superoxide dismutase (SOD) activity. Activities of SOD, CAT and GST as well as H 2 O 2 levels were not significantly affected in CAP and CAP plus MHC-treated rats whereas GSH level and 5¢ NTD activity were markedly decreased in CAP plus MHC-treated rats. Significant increase in testicular alkaline phosphatase activity, lipid peroxidation and sperm abnormalities were accompanied by reduction in epididymal sperm number, sperm motility and livedead ratio in all treatment groups whereas aminotransferase activities were unaffected. Treatment-related degeneration of the testes was evident in all treated animals. In summary, while MHC-induced testicular toxicity via oxidative stress, CAP did not and their combination is implicated in reproductive dysfunction within the time course of our investigation.

Antibiotics are widely used and their abuse has caused ecological hazard. Recently, pollution from pharmaceuticals and personal care products (PPCPs) has aroused great concern among governments and researchers. In order to elucidate the correlations among molecular structure, transmembrane distribution and toxicological effects of different kinds of antibiotics, zebrafish (Danio rerio) embryos and larvae were exposed to two structurally different antibiotics, kanamycin (KAN) and chloramphenicol (CAP). The membrane distribution and toxicological effects of these antibiotics were investigated. The association of KAN with the embryos fitted a general Langmuir isotherm and was attributed to electrostatic attraction and hydrogen bond formation. The saturation number of KAN is 252 +/- 13 nmol per embryo and the adsorption constant (5.24 +/- 0.05) x 10(3) L mol(-1). The interaction of CAP with the embryos conformed to a general model of partitioning behavior with the partition coefficient being 14.20 +/- 0.94 microL per embryo, and was attributed to hydrophobic effects. More than 89% of the adsorbed KAN was located on the outer surface of the embryonic chorion, but over 80% of the adsorbed CAP entered the internal matrix. High antibiotic concentrations were lethal to most embryos, while low concentrations were teratogenic. KAN and CAP had different transmembrane distribution and their toxicities differed in character. KAN mainly accumulated on the outer membrane caused e.g. axial malformation (AM). In contrast, CAP readily went through the membrane into the cytoplasm and caused e.g. serious pericardial edema (PE), yolk sac edema (YSE) and hemagglutination (HE). The new method could be useful for evaluating the interactions of toxins with membranes and elucidating the mechanisms of cytotoxicity.

Background: Isotretinoin (ISO) belongs to a family of drugs called retinoids. It is the most effective drug prescribed by dermatologists for the treatment of the inflammatory disease, acne vulgaris. A significant barrier to the use of ISO has worries regarding its adverse effect profile. Despite the well-recognized reproductive toxicity and teratogenicity in females, there is no warning related to the use by male patients in the medication prospectus. Current data on the effects on human male fertility is contradictory and inconclusive. Objectives: This study was undertaken to investigate the potential effects of ISO oral doses in the Sprague–Dawley male rat germ cells using the sperm morphology assay. Also, the serum levels of the follicle-stimulating hormone (FSH), luteinizing hormone (LH), and testosterone were measured. Methods: The rat groups were given varying ISO doses via gastric gavage for seven consecutive days. The epididymis sperm specimens were microscopically examined for the following reproductive toxicity parameters: sperm concentration, examined viability, motility, and morphology. The serum FSH, LH, and testosterone levels were measured by using the corresponding enzyme-linked immunosorbent assay (ELISA) kit. The data were analyzed statistically by one-way analysis of variance (ANOVA) followed by the Tukey test at P ≤ 0.05 significance level. Results: The results indicated that the drug did not significantly increase the sex hormone levels but notably affected both the sperm quantity and quality. Conclusion: These observations suggest that ISO was reprotoxic, and future therapies should be further reassessed.
